The abnormal expression and polymerization of ACTB and the resulting changes to the cytoskeleton are revealed to be associated with the invasiveness and metastasis of cancers. Diseases associated with ACTB include Baraitser-Winter Syndrome 1 and Dystonia-Deafness Syndrome 1.<br><br>Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti beta-actin antibody was precoated onto the 96-well plate. The biotin conjugated anti beta-actin antibody was used as the detection antibody. The standards and pilot samples were added to the wells subsequently. After incubation, unbound conjugates were removed by wash buffer. Then, biotinylated detection antibody was added to bind with beta-actin conjugated on coated antibody. After washing off unbound conjugates, HRP-Streptavidin was added. After a third washing, TMB substrates were added to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that turned yellow after adding a stop solution. Read the O.D. absorbance at 450nm in a microplate reader. The concentration of beta-actin in the sample was calculated by drawing a standard curve. Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for F-actin has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any F-actin present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for F-actin is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of F-actin bound in the initial step. The color development is stopped and the intensity of the color is measured.

Actin can be resolved on the basis of its isoelectric points into three distinctive components: alpha, beta, and gamma in order of increasing isoelectric point. Anti-muscle specific actin recognizes alpha and gamma isotypes of all muscle groups. Non-muscle cells such as vascular endothelial cells and connective tissues are non-reactive. Also, neoplastic cells of non-muscle-derived tissue such as carcinomas, melanomas, and lymphomas are negative. Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for IL-6 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any IL-6 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for IL-6 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of IL-6 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Principle of the Assay: GACT ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-GACT antibody and an GACT-HRP conjugate. The assay sample and buffer are incubated together with GACT-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GACT concentration since GACT from samples and GACT-HRP conjugate compete for the anti-GACT antibody binding site. Since the number of sites is limited, as more sites are occupied by GACT from the sample, fewer sites are left to bind GACT-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GACT concentration in each sample is interpolated from this standard curve.<br><br>Intended Uses: This GACT ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human GACT. Principle of the Assay: As mentioned above, this kit utilizes the Double Antibody Sandwich ELISA technique. The pre-coated antibody is an anti-Mouse alpha-SMA monoclonal antibody, while the detection antibody is a biotinylated polyclonal antibody. Samples and biotinylated antibodies are added into ELISA plate wells and washed out with PBS or TBS after their respective additions to the wells. Then Avidin-peroxidase conjugates are added to the wells in after. TMB substrate is used for coloration after the enzyme conjugate has already been thoroughly washed out of the wells by PBS or TBS. TMB reacts to form a blue product from the peroxidase activity, and finally turns to yellow after addition of the stop solution (Color Reagent C).
