Intended Uses: This CD15 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of HumanCD15. This ELISA kit for research use only!

Principle of the Assay: CD15 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CD15 antibody and an CD15-HRP conjugate. The assay sample and buffer are incubated together with CD15-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CD15 concentration since CD15 from samples and CD15-HRP conjugate compete for the anti-CD15 antibody binding site. Since the number of sites is limited, as more sites are occupied by CD15 from the sample, fewer sites are left to bind CD15-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CD15 concentration in each sample is interpolated from this standard curve.

NCBI Description

CD160 is an 27 kDa glycoprotein which was initially identified with the monoclonal antibody BY55. Its expression is tightly associated with peripheral blood NK cells and CD8 T lymphocytes with cytolytic effector activity. The cDNA sequence of CD160 predicts a cysteine-rich, glycosylphosphatidylinositol-anchored protein of 181 amino acids with a single Ig-like domain weakly homologous to KIR2DL4 molecule. CD160 is expressed at the cell surface as a tightly disulfide-linked multimer. RNA blot analysis revealed CD160 mRNAs of 1.5 and 1.6 kb whose expression was highly restricted to circulating NK and T cells, spleen and small intestine. Within NK cells CD160 is expressed by CD56dimCD16+ cells whereas among circulating T cells its expression is mainly restricted to TCRgd bearing cells and to TCRab+CD8brightCD95+CD56+CD28-CD27-cells. In tissues, CD160 is expressed on all intestinal intraepithelial lymphocytes. CD160 shows a broad specificity for binding to both classical and nonclassical MHC class I molecules.
