Mouse Cbp/p300-interacting transactivator 2 (CITED2) ELISA Kit
Principle of the Assay||CITED2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CITED2 antibody and an CITED2-HRP conjugate. The assay sample and buffer are incubated together with CITED2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CITED2 concentration since CITED2 from samples and CITED2-HRP conjugate compete for the anti-CITED2 antibody binding site. Since the number of sites is limited, as more sites are occupied by CITED2 from the sample, fewer sites are left to bind CITED2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CITED2 concentration in each sample is interpolated from this standard curve.

Associates with chromatin to the PITX2 P1 promoter region. Defects in CITED2 are a cause of ventricular septal defect type 2 (VSD2). VSD2 is a common form of congenital cardiovascular anomaly that may occur alone or in combination with other cardiac malformations. It can affect any portion of the ventricular septum, resulting in abnormal communications between the two lower chambers of the heart. Classification is based on location of the communication, such as perimembranous, inlet, outlet (infundibular), central muscular, marginal muscular, or apical muscular defect. Large defects that go unrepaired may give rise to cardiac enlargement, congestive heart failure, pulmonary hypertension, Eisenmenger's syndrome, delayed fetal brain development, arrhythmias, and even sudden cardiac death. Defects in CITED2 are a cause of atrial septal defect type 8 (ASD8). ASD8 is a congenital heart malformation characterized by incomplete closure of the wall between the atria resulting in blood flow from the left to the right atria. Belongs to the CITED family. 2 isoforms of the human protein are produced by alternative splicing.
