Mouse Calcium/calmodulin-dependent protein kinase kinase 2 (CAMKK2) ELISA Kit
Principle of the Assay: CAMKK2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CAMKK2 antibody and an CAMKK2-HRP conjugate. The assay sample and buffer are incubated together with CAMKK2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CAMKK2 concentration since CAMKK2 from samples and CAMKK2-HRP conjugate compete for the anti-CAMKK2 antibody binding site. Since the number of sites is limited, as more sites are occupied by CAMKK2 from the sample, fewer sites are left to bind CAMKK2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CAMKK2 concentration in each sample is interpolated from this standard curve.

The product of this gene belongs to the Serine/Threonine protein kinase family, and to the Ca(2+)/calmodulin-dependent protein kinase subfamily. The major isoform of this gene plays a role in the calcium/calmodulin-dependent (CaM) kinase cascade by phosphorylating the downstream kinases CaMK1 and CaMK4. Protein products of this gene also phosphorylate AMP-activated protein kinase (AMPK). This gene has its strongest expression in the brain and influences signalling cascades involved with learning and memory, neuronal differentiation and migration, neurite outgrowth, and synapse formation. Alternative splicing results in multiple transcript variants encoding distinct isoforms.
