Mouse ATP-binding cassette sub-family D member 4 (ABCD4) ELISA Kit
Principle of the Assay: ABCD4 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ABCD4 antibody and an ABCD4-HRP conjugate. The assay sample and buffer are incubated together with ABCD4-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ABCD4 concentration since ABCD4 from samples and ABCD4-HRP conjugate compete for the anti-ABCD4 antibody binding site. Since the number of sites is limited, as more sites are occupied by ABCD4 from the sample, fewer sites are left to bind ABCD4-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ABCD4 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a member of the superfamily of ATP-binding cassette (ABC) transporters. ABC proteins transport various molecules across extra- and intra-cellular membranes. ABC genes are divided into seven distinct subfamilies (ABC1, MDR/TAP, MRP, ALD, OABP, GCN20, White). This protein is a member of the ALD subfamily, which is involved in peroxisomal import of fatty acids and/or fatty acyl-CoAs in the organelle. All known peroxisomal ABC transporters are half transporters which require a partner half transporter molecule to form a functional homodimeric or heterodimeric transporter. The function of this peroxisomal membrane protein is unknown. However, it is speculated that it may function as a heterodimer for another peroxisomal ABC transporter and, therefore, may modify the adrenoleukodystrophy phenotype. It may also play a role in the process of peroxisome biogenesis.
