Degradation of mRNA is a critical aspect of gene expression that occurs via the exoribonuclease. Exoribonuclease 2 (XRN2) is the human homologue of the Saccharomyces cerevisiae RAT1, which functions as a nuclear 5' to 3' exoribonuclease and is essential for mRNA turnover and cell viability. XRN2 also processes rRNAs and small nucleolar RNAs (snoRNAs) in the nucleus. XRN2 moves along with RNA polymerase II and gains access to the nascent RNA transcript after the endonucleolytic cleavage at the poly (A) site or at a second cotranscriptional cleavage site (CoTC). CoTC is an autocatalytic RNA structure that undergoes rapid self-cleavage and acts as a precursor to termination by presenting a free RNA 5' end to be recognized by XRN2. XRN2 then travels in a 5'-3' direction like a guided torpedo and facilitates the dissociation of the RNA polymerase elongation complex.

NCBI Description

This gene shares similarity with the mouse Dhm1 and the yeast dhp1 gene. The yeast gene is involved in homologous recombination and RNA metabolism, such as RNA synthesis and RNA trafficking. Complementation studies show that Dhm1 has a similar function in mouse as dhp1. The function of the human gene has not yet been determined. Transcript variants encoding different isoforms have been noted for this gene; however, their full-length nature is not known. [provided by RefSeq, Jul 2008]

XRN2: Possesses 5'->3' exoribonuclease activity. May promote the termination of transcription by RNA polymerase II. During transcription termination, cleavage at the polyadenylation site liberates a 5' fragment which is subsequently processed to form the mature mRNA and a 3' fragment which remains attached to the elongating polymerase. The processive degradation of this 3' fragment by this protein may promote termination of transcription. Belongs to the 5'-3' exonuclease family. XRN2/RAT1 subfamily. 2 isoforms of the human protein are produced by alternative splicing.
