Clone Number
TMS715
Specificity
It recognizes a protein of 36kDa, identified as Thymidylate Synthase (TS) (EC 2.1.1.45). TS converts deoxyuridine monophosphate (dUMP) to deoxythymidine monophosphate (dTMP), which is essential for DNA biosynthesis. TS is also a critical target for the fluoropyrimidines, an important group of antineoplastic drugs that are widely used in the treatment of solid tumors. Both 5-FU and fluorodeoxyuridine are converted in tumor cells to FdUMP which inactivates TS by formation of a ternary covalent complex in the presence of the folate cofactor 5,10-methylenetetrahydrofolate. Expression of TS protein is associated with response to 5-fluorouracil (5-FU) in human colorectal, gastric, head and neck, and breast carcinomas.

NCBI Description

Thymidylate synthase catalyzes the methylation of deoxyuridylate to deoxythymidylate using 5,10-methylenetetrahydrofolate (methylene-THF) as a cofactor. This function maintains the dTMP (thymidine-5-prime monophosphate) pool critical for DNA replication and repair. The enzyme has been of interest as a target for cancer chemotherapeutic agents. It is considered to be the primary site of action for 5-fluorouracil, 5-fluoro-2-prime-deoxyuridine, and some folate analogs. Expression of this gene and that of a naturally occuring antisense transcript rTSalpha (GeneID:55556) vary inversely when cell-growth progresses from late-log to plateau phase. Uniprot Description

TYMS: an enzyme in the thymidylate synthase family. Polymorphisms serve as prognostic markers for determining modality of treatment in certain cancers. Genotyping helps predict toxicity to 5-FU-based chemotherapy in patients with colorectal cancer and the response to platinum-based chemotherapy in advanced NSCLC. A common polymorphism is a determinant of red blood cell folate and homocysteine concentrations. Homodimer.

Product Notes

The TYMS tyms (Catalog #AAA438476) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The Thymidylate Synthase (5-FU Resistance Marker) Mouse Monoclonal Antibody reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's Thymidylate Synthase can be used in a range of immunoassay formats including, but not limited to, Flow Cytometry (FC/FACS), Immunofluorescence (IF), Immunohistochemistry (IHC) Formalin. Flow Cytometry (0.5-1ug/million cells in 0.1ml) Immunofluorescence (0.5-1ug/ml) Immunohistochemistry (Formalin-fixed) (1-2ug/ml for 30 minutes at RT) (Staining of formalin-fixed tissues is enhanced by boiling tissue sections in 10mM Citrate Buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 minutes). Researchers should empirically determine the suitability of the TYMS tyms for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Thymidylate Synthase, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
