alpha-synuclein (alpha-syn) is a 14-19 kDa phosphoprotein that is primarily localize to the presynaptic terminals of mature neurons, where it is involved in synaptic function and plasticity. alpha-syn has drawn intense interest ever since the late 1990s, when the first alpha-synuclein missense mutation was identified as a cause of familial Parkinson's disease (PD). Aggregated and hyper-phosphorylated forms of alpha-syn protein are the pathological hallmark of Lewy body disease, which includes Parkinson's disease (PD), diffuse Lewy body disease (DLBD), and Lewy body variant of Alzheimer's disease (LBV). This antibody can recognize all the isoforms of alpha-syn.

Alpha-synuclein is a member of the synuclein family, which also includes beta- and gamma-synuclein. Synucleins are abundantly expressed in the brain and alpha- and beta-synuclein inhibit phospholipase D2 selectively. SNCA may serve to integrate presynaptic signaling and membrane trafficking. Defects in SNCA have been implicated in the pathogenesis of Parkinson disease. SNCA peptides are a major component of amyloid plaques in the brains of patients with Alzheimer's disease. Alternatively spliced transcripts encoding different isoforms have been identified for this gene. [provided by RefSeq, Feb 2016]
