Entrez Summary: Cell growth is a fundamental biological process whereby cells accumulate mass and increase in size. The mammalian TOR (mTOR) pathway regulates growth by coordinating energy and nutrient signals with growth factor-derived signals. mTOR is a large protein kinase with two different complexes. One complex contains mTOR, GbetaL and raptor, which is a target of rapamycin. The other complex, insensitive to rapamycin, includes mTOR, GbetaL, Sin1 and rictor. The mTOR-rictor complex phosphorylates Ser473 of Akt/PKB in vitro. This phosphorylation is essential for full Akt/PKB activation. Furthermore, an siRNA knockdown of rictor inhibits Ser473 phosphorylation in 3T3-L1 adipocytes. This complex has also been shown to phosphorylate the rapamycin-resistant mutants of S6K1, another effector of mTOR.

NCBI Description

RICTOR and MTOR (FRAP1; MIM 601231) are components of a protein complex that integrates nutrient- and growth factor-derived signals to regulate cell growth (Sarbassov et al., 2004 [PubMed 15268862]).[supplied by OMIM, Mar 2008]

Uniprot Description

RICTOR: a regulatory binding partner of the kinase mTOR. Part of a rapamycin-insensitive and raptor-independent pathway that regulates the cytoskeleton. Rictor interacts with Cullin1-Rbx1 to form an E3 ubiquitin ligase complex, and promotes ubiquitination and degradation of SGK1. Shares homology with pianissimo from D. discoidieum, STE20p from S. pombe, and AVO3p from S. cerevisiae.

Protein type: Adaptor/scaffold; Cell cycle regulation; Ubiquitin conjugating system

Chromosomal Location of Human Ortholog: 5p13.1

Cellular Component: cytosol; TORC2 complex

Molecular Function: enzyme activator activity; protein binding; protein kinase binding

Biological Process: actin cytoskeleton reorganization; embryonic development; establishment and/or maintenance of actin cytoskeleton polarity; establishment of cell polarity; peptidyl-serine phosphorylation; phosphoinositide-mediated signaling; positive regulation of protein kinase B signaling cascade; positive regulation of TOR signaling pathway; regulation of actin cytoskeleton organization and biogenesis; regulation of phosphorylation; regulation of protein kinase B signaling cascade; T cell costimulation

Product Notes

The RICTOR rictor (Catalog #AAA476060) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The Anti-RICTOR Mouse mAb reacts with Human, Monkey, Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's RICTOR can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Researchers should empirically determine the suitability of the RICTOR rictor for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "RICTOR, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
