Related Product Information for anti-PLCG1 antibody
Phosphoinositide-specific phospholipase C (PLC) plays a significant role in transmembrane signaling. In response to extracellular stimuli such as hormones, growth factors, and neurotransmitters, PLC hydrolyzes phosphatidylinositol 4,5-bisphosphate (PIP2) to generate two secondary messengers: inositol 1,4,5-triphosphate (IP3) and diacylglycerol (DAG). At least four families of PLCs have been identified: PLC-beta, PLC-gamma, PLC-delta, and PLC-epsilon. Phosphorylation is one of the key mechanisms that regulate the activity of PLC. PLC-delta is activated by both receptor and nonreceptor tyrosine kinases. PLC-gamma-1 forms a complex with EGF and PDGF receptors, which leads to phosphorylation at tyrosine 771, 783, and 1245. In addition, antigen receptor-induced activation of PLC-gamma-1 leads to phosphorylation at both Tyr-775 and Tyr-783. These two sites are equally important for activation of enzymatic activity.

NCBI Description

The protein encoded by this gene catalyzes the formation of inositol 1,4,5-trisphosphate and diacylglycerol from phosphatidylinositol 4,5-bisphosphate. This reaction uses calcium as a cofactor and plays an important role in the intracellular transduction of receptor-mediated tyrosine kinase activators. For example, when activated by SRC, the encoded protein causes the Ras guanine nucleotide exchange factor RasGRP1 to translocate to the Golgi, where it activates Ras. Also, this protein has been shown to be a major substrate for heparin-binding growth factor 1 (acidic fibroblast growth factor)-activated tyrosine kinase. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq]

Uniprot Description

Function: Plays a role in actin reorganization and cell migration. The production of the second messenger molecules diacylglycerol (DAG) and inositol 1,4,5-trisphosphate (IP3) is mediated by activated phosphatidylinositol-specific phospholipase C enzymes. Major substrate for heparin-binding growth factor 1 (acidic fibroblast growth factor)-activated tyrosine kinase. Ref.17

Catalytic activity: 1-phosphatidyl-1D-myo-inositol 4,5-bisphosphate + H2O = 1D-myo-inositol 1,4,5-trisphosphate + diacylglycerol.

Cofactor: Calcium.

Subunit structure: Interacts with AGAP2 via its SH3 domain

By similarity. Interacts with phosphorylated LAT upon TCR activation. Interacts with the Pro-rich domain of TNK1 via its SH3 domain. Associates with BLNK, VAV1, GRB2 and NCK1 in a B-cell antigen receptor-dependent fashion. Interacts with CBLB in activated T-cells; which inhibits phosphorylation. Interacts with SHB. Interacts via its SH3 domain with the Arg/Gly-rich-flanked Pro-rich domains of KHDRBS1/SAM68. This interaction is selectively regulated by arginine methylation of KHDRBS1/SAM68. Interacts with INPP5D/SHIP1, THEMIS and CLNK

By similarity. Interacts with RALGPS1. Interacts (via SH3 domain) with HEV ORF3 protein. Interacts (via the SH2 domains) with VIL1 (phosphorylated at C-terminus tyrosine phosphorylation sites). Ref.6 Ref.7 Ref.8 Ref.9 Ref.10 Ref.13 Ref.17

Subcellular location: Cell projection › lamellipodium. Cell projection › ruffle. Note: Rapidly redistributed to ruffles and lamellipodia structures in response to epidermal growth factor (EGF) treatment. Ref.17

Domain: The SH3 domain mediates interaction with CLNK

By similarity. The SH3 domain also mediates interaction with RALGPS1. Ref.26

Post-translational modification: The receptor-mediated activation of PLC-gamma-1 and PLC-gamma-2 involves their phosphorylation by tyrosine kinases in response to ligation of a variety of growth factor receptors and immune system receptors. May be dephosphorylated by PTPRJ. Ref.11 Ref.12 Ref.14 Ref.15 Ref.16 Ref.18 Ref.19 Ref.20 Ref.21 Ref.22 Ref.23 Ref.24Ubiquitinated by CBLB in activated T-cells

Product Notes

The PLCG1 plcg1 (Catalog #AAA470026) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The Anti-PLC-gamma1 (N-terminal region) reacts with Human, Rat, Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's PLCg1 (N-terminal Region) can be used in a range of immunoassay formats including, but not limited to, ELISA, Western Blot, Immunocytochemistry, Immunoprecipitation. ELISA: 1:2000 Immunocytochemistry: 1:100 Immunoprecipitation: 1-5ml Western Blot: 1:1000. Researchers should empirically determine the suitability of the PLCG1 plcg1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "PLCg1 (N-terminal Region), Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
