Specificity
Recognizes a 38kD nucleolar protein (NO38) in Xenopus (this protein has also been termed B23, nucleophosmin, numatrin in other species). Stains specifically the granular component of the nucleolus and small nucleolar dots in interphase cells. During mitosis the protein is localized accumulated around the condensed chromosomes. Species Crossreactivity: Xenopus laevis, Xenopus borealis, Pleurodeles walt, Triturus cristatus; XLKE-A6 cells; chicken.

Application Notes
Suitable for use in Western Blot, Immunoprecipitation, Immunofluorescence and Immunohistochemistry.
Dilution: Western Blot: 1:1000-1:2000 (ECL method) Nitrocellulose sheet are better then nylon sheets.
Immunohistochemistry: 1:300-1:500; (frozen sections, also after fixation with paraformaldehyde).
Immunoprecipitation: from Xenopus oocyte extracts nucleoplasmin-a 30kD protein-is co-precipitated, which shows high sequence homology to NO38.

Nucleolar fraction prepared from Xenopus laevis oocytes.
Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

MCRS1: Modulates the transcription repressor activity of DAXX by recruiting it to the nucleolus. As part of the NSL complex it may be involved in acetylation of nucleosomal histone H4 on several lysine residues. Putative regulatory component of the chromatin remodeling INO80 complex which is involved in transcriptional regulation, DNA replication and probably DNA repair. May also be an inhibitor of TERT telomerase activity. 3 isoforms of the human protein are produced by alternative splicing.
