Application Notes
Suitable for use in Direct ELISA, Western Blot and Immunohistochemistry.
Dilution: Direct ELISA: 0.5-1ug/ml; The detection limit for recombinant mouse Netrin G2a is ~3ng/well.
Western Blot: 1-2ug/ml. Using a colorimetric detection system, the detection limit for recombinant mouse Netrin-G2a is ~10ng/lane under non-reducing and reducing conditions. Chemiluminescent detection will increase sensitivity by 5 to 50 fold.
Immunohistochemistry: 25ug/ml detects Netrin G2a in frozen normal mouse brain tissue sections.

Mouse myeloma with B cells obtained from a rat immunized with purified, NS0-derived, recombinant mouse Netrin G2a.
Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Mouse Netrin G2a is a member of a GPI-linked Netrin subfamily that is distantly related to classical Netrins. Netrin G2a is approximately 485aa in length. It contains an N-terminal laminin-related region plus a heparin-binding C-terminus that ends in a GPI-linkage. The laminin-related region shows a globular type VI domain followed by three variable-length EGF-like domains. Alternate splicing generates two larger isoforms. Amino acid insertions of 59aa and 34aa between EGF-like motifs 1 and 2 generate Netrin G2b and Netrin G2c, respectively. Mature mouse Netrin G2a shares 96% and 99% amino acid identity with human Netrin G2 and the equivalent region in rat Netrin G2b, respectively.

NCBI Description

The protein encoded by this gene belongs to a subclass of the netrin family called netrin-G proteins. Unlike classic netrins, which act as diffusible chemoattractants, netrin-Gs are glycosylphosphatidylinositol-anchored membrane proteins that interact with specific transmembrane proteins. In mouse, this gene is preferentially expressed in the cerebral cortex, habenular nucleus and superior colliculus. Knockout mutant mice display a lack of behavioral startle in response to acoustic stimuli. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Apr 2015]
