In a sandwich ELISA (assuming 100ul/well), a concentration of 2.0-4.0 ug/ml of this antibody will detect at least 2000 pg/ml of recombinant human MIP-1alpha when used with an appropriate secondary reagent as the detection antibody.

Produced in BALB/c mice using highly pure (>98%) recombinant human MIP 1alpha as the immunizing antigen. This IgG1K antibody was purified from cell culture by Protein G affinity chromatography.

Both MIP-1alpha and MIP-1beta are structurally and functionally related CC chemokines. They participate in the host response to invading bacterial, viral, parasite and fungal pathogens by regulating the trafficking and activation state of selected subgroups of inflammatory cells e.g. macrophages, lymphocytes and NK cells. While both MIP-1alpha and MIP-1beta exert similar effects on monocytes their effect on lymphocytes differ; with MIP-1alpha selectively attracting CD8+ lymphocytes and MIP-1beta selectively attracting CD4+ lymphocytes. Additionally, MIP-1alpha and MIP-1beta have also been shown to be potent chemoattractants for B cells, eosinophils and dendritic cells. Both human and murine MIP-1alpha and MIP-1beta are active on human and murine hematopoietic cells. Recombinant human MIP-1alpha is a 7.8 kDa protein containing 69 amino acid residues, including the four highly conserved cysteine residues present in CC chemokines.

This locus represents a small inducible cytokine. The encoded protein, also known as macrophage inflammatory protein 1 alpha, plays a role in inflammatory responses through binding to the receptors CCR1, CCR4 and CCR5. Polymorphisms at this locus may be associated with both resistance and susceptibility to infection by human immunodeficiency virus type 1.[provided by RefSeq, Sep 2010]

Uniprot Description

Function: Monokine with inflammatory and chemokinetic properties. Binds to CCR1, CCR4 and CCR5. One of the major HIV-suppressive factors produced by CD8+ T-cells. Recombinant MIP-1-alpha induces a dose-dependent inhibition of different strains of HIV-1, HIV-2, and simian immunodeficiency virus (SIV). Ref.8

Subunit structure: Self-associates. Also heterodimer of MIP-1-alpha(4-69) and MIP-1-beta(3-69). Ref.11

Subcellular location: Secreted.

Induction: By TPA or PHA (TPA = 12-O-tetradecanoyl phorbol-13 acetate (tumor promoter); PHA = phytohemagglutinin (T-cell mitogen)).

Post-translational modification: N-terminal processed form LD78-alpha(4-69) is produced by proteolytic cleavage after secretion from HTLV1-transformed T-cells.

Sequence similarities: Belongs to the intercrine beta (chemokine CC) family.
