Flow Cytometry (FC/FACS)
Application Notes
Suitable for use in Flow Cytometry.
Dilution: Flow Cytometry: 10ul labels up to 1x10e6 cells. Validated for use on HUVEC cells.
Flow Cytometry Protocol: 1. Cells may be Fc-blocked with 1ug of human IgG/10e5 cells for 15 minutes at RT. Do not wash excess blocking IgG from this reaction.
2. After blocking, add 10ul of L9375-01B to up to 1x10e6 cells and incubate for 30 minutes at RT.
3. Remove unbound antibody by washing the cells twice in Flow Cytometry Staining Buffer (F4568-10). Note that whole blood requires a RBC lysis step at this point using Flow Cytometry Human Lyse Buffer.
4. Resuspend the cells in Flow Cytometry Staining Buffer for final flow cytometric analysis. As a control for this analysis, cells in a separate tube should be treated with APC-labeled mouse IgG1 antibody.
Immunogen
Recombinant protein corresponding to aa20-238 from human LYVE1 expressed in E. coli (Q9Y5Y7).
Preparation and Storage
May be stored at 4 degree C before opening. DO NOT FREEZE! Stable at 4 degree C as an undiluted liquid. Stable for at 12 months at 4 degree C. Freezing allophycocyanin conjugates will result in a substantial loss of activity. Allophycocyanin conjugates are sensitive to light.

LYVE1, also named extracellular link domain containing 1, is a 60-70kD type I transmembrane protein that functions as a receptor for both soluble and immobilized hyaluronan (HA). It shares 41% aa sequence homology with CD44, another known receptor for HA. LYVE1 expression is mostly restricted to lymphatic vessels. LYVE1 is a useful molecular marker for lymphatic endothelial cells.

Function: Ligand-specific transporter trafficking between intracellular organelles (TGN) and the plasma membrane. Plays a role in autocrine regulation of cell growth mediated by growth regulators containing cell surface retention sequence binding (CRS). May act as a hyaluronan (HA) transporter, either mediating its uptake for catabolism within lymphatic endothelial cells themselves, or its transport into the lumen of afferent lymphatic vessels for subsequent re-uptake and degradation in lymph nodes. Ref.1

Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "LYVE1, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening.
