Application Notes
The antibody has been tested by ELISA, Western blot analysis to assure specificity and reactivity. Since application varies, however, each investigation should be titrated by the reagent to obtain optimal results. Recommended starting dilution is 1:1000.
Immunogen
Anti-human ICT1 mAb, clone PAT1E9A, is derived from hybridization of mouse F0 myeloma cells with spleen cells from BALB/c mice immunized with a recombinant human ICT1 protein 30-206 amino acids purified from E Coli.
Preparation and Storage
For periods up to 1 month store at 4 degree C, for longer periods of time, store at -20 degree C. Prevent freeze thaw cycles.
12 months at -20 degree C. 1 month at 4 degree C.

Introduction: Immature Colon Carcinoma Transcript 1 (ICT1) functions as a codon-independent translation release factor that has lost all stop codon specificity and leads the termination of translation in mitochondrion in case of abortive elongation. The mature colon epithelium has 3 differentiated cell types which arise from a multipotent stem cell. ICT1 is involved in the hydrolysis of peptidyl-tRNAs which have been prematurely terminated and consequently in the recycling of stalled mitochondrial ribosomes. Digression from the normal maturation pathway by neoplastic transformation is assumed to originate in stem cells or their early descendants.

NCBI Description

The protein encoded by this gene is a peptidyl-tRNA hydrolase and a vital component of the large mitochondrial ribosome. The encoded protein serves as a ribosome release factor for this ribosome, which translates mitochondrial genes. This protein may be responsible for degrading prematurely-terminated polypeptides and for reusing stalled ribosomes. Two transcript variants encoding different isoforms have been found for this gene.
