Rat anti-Mouse IL2R gamma C Monoclonal Antibody | anti-IL2RgammaC antibody
Anti-IL2R gamma C [3E12]
Immunofluorescence (IF)
(Immunofluorescence staining of fixed mouse splenocytes with anti-IL2R gamma C antibody 3E12 (MBS488670) Immunofluorescence analysis of paraformaldehyde fixed mouse splenocytes on Shi-fix coverslips stained with the chimeric rabbit IgG version of 3E12 (MBS488670) at 10ug/ml for 1h followed by Alexa Fluor 488 secondary antibody (2ug/ml), showing membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom MBS488670, DAPI, merged channels and an isotype control. The isotype control was an unknown specificity antibody (MBS488140) followed by staining with Alexa Fluor 488 secondary antibody.)
This antibody was raised by immunising Lewis rats with transfected rat myeloma cell line YB2/0, expressing murine cytoplasmic-tailless IL-2R common gamma-chain (YB2/0-myt).
Note on publication: Describes the original generation of this antibody, and its use in determining murine common gamma-chain lymphoid cell distribution.
NCBI and Uniprot Product Information
NCBI Description
This gene encodes a transmembrane protein that is a common subunit of several interleukin receptor complexes. These receptors are comprised of alpha and beta subunits in addition to this gamma subunit. Signalling through this pathway in important in immune cell differentiation and function. Alternative splicing results in multiple transcript variants. [provided by RefSeq, May 2015]
Uniprot Description
IL2RG: Common subunit for the receptors for a variety of interleukins. Defects in IL2RG are the cause of severe combined immunodeficiency X-linked T-cell-negative/B-cell-positive/NK-cell- negative (XSCID); also known as agammaglobulinemia Swiss type. A form of severe combined immunodeficiency (SCID), a genetically and clinically heterogeneous group of rare congenital disorders characterized by impairment of both humoral and cell- mediated immunity, leukopenia, and low or absent antibody levels. Patients present in infancy recurrent, persistent infections by opportunistic organisms. The common characteristic of all types of SCID is absence of T-cell-mediated cellular immunity due to a defect in T-cell development. Defects in IL2RG are the cause of X-linked combined immunodeficiency (XCID). XCID is a less severe form of X-linked immunodeficiency with a less severe degree of deficiency in cellular and humoral immunity than that seen in XSCID. Belongs to the type I cytokine receptor family. Type 5 subfamily.
Protein type: Receptor, cytokine; Membrane protein, integral
Cellular Component: cell surface; membrane; integral to membrane; external side of plasma membrane
Molecular Function: hematopoietin/interferon-class (D200-domain) cytokine receptor activity; interleukin-7 binding; interleukin-2 binding
Biological Process: positive regulation of T cell differentiation in the thymus; positive regulation of lymphocyte differentiation; positive regulation of CD4-positive, CD25-positive, alpha-beta regulatory T cell differentiation; regulation of gene expression; positive regulation of B cell differentiation

Product Notes
The IL2RgammaC il2rg (Catalog #AAA488671) is an Antibody produced from Rat and is intended for research purposes only. The product is available for immediate purchase. The Anti-IL2R gamma C [3E12] reacts with Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's IL2R gamma C can be used in a range of immunoassay formats including, but not limited to, Flow Cytometry (FC/FACS), Immunoprecipitation (IP), Blocking (BL). This antibody has been used in flow cytometric analysis of common gamma-chain surface expression by IL-7-stimulated T cells (Goh et al, 2017) and COS7 monkey kidney cells (Olosz et al, 2002). This antibody has also been used to immunoprecipiate murine common gamma-chain-containing complexes from COS7 cell extracts (Olosz et al, 2002). Additionally, as this antibody is conformationally sensitive, it has been used to confirm the correct folding of soluble murine common gamma chain mutants (Meibetaner et al, 2001). This antagonistic antibody is most effective at inhibiting IL-4 bioactivity (Malek et al, 1998). Treatment of recipient mice with this antibody, along with another common gamma-chain-blocking MAb (4G3), significantly prolongs islet allograft survival (Li et al, 2000). Indeed, blocking the common gamma chain with this antibody has been shown to induce apoptotic cell death of BAF-B03 cells and primary anti-CD3 activated T lymphoblasts in vitro, and of alloreactive T cells in vivo (Li et al, 2000). Researchers should empirically determine the suitability of the IL2RgammaC il2rg for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "IL2R gamma C, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
