Specificity
Immunoglobulins are four-chain, Y-shaped, monomeric structures comprised of two identical heavy chains and two identical light chains held together through interchain disulfide bonds. The chains form two domains, the Fab (antigen binding) fragment and the Fc (constant) fragment. Immunoglobulin D (IgD) exists as a monomer with light chains. It plays a biological role as a transmembrane receptor molecule, co-expressed with IgM on the surface of mature/naive B cells. In particular, it is found on spleen B cell surfaces. Compared to IgM, IgD exists in much lower numbers and is not expressed on immature B cells. IgD surface expression on B cells is regulated in part by IL-27. In mice, the inhibition of this immunoglobulin isotype does not cause a significant change to the immune system.

Uniprot Description

IGHD: IgD is the major antigen receptor isotype on the surface of most peripheral B-cells, where it is coexpressed with IgM. The membrane-bound IgD (mIgD) induces the phosphorylation of CD79A and CD79B by the Src family of protein tyrosine kinases. Soluble IgD (sIgD) concentration in serum below those of IgG, IgA, and IgM but much higher than that of IgE. IgM and IgD molecules present on B cells have identical V regions and antigen-binding sites. After the antigen binds to the B-cell receptor, the secreted form sIgD is shut off. IgD is a potent inducer of TNF, IL1B, and IL1RN. IgD also induces release of IL6, IL10, and LIF from peripheral blood mononuclear cells. Monocytes seem to be the main producers of cytokines in vitro in the presence of IgD. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes

The IGHD ighd (Catalog #AAA4380996) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The IgD (Immunoglobulin Delta Heavy Chain) (B-Cell Marker) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's IgD (Immunoglobulin Delta Heavy Chain) can be used in a range of immunoassay formats including, but not limited to, Flow Cytometry (FC/FACS), Immunofluorescence (IF), Immunohistochemistry (IHC) Paraffin. FC/FACS: 0.5-1ug/million cells IF: 0.5-1ug/ml Immunohistochemistry (Formalin-fixed) (0.5-1ug/ml for 30 minutes at RT) (Staining of formalin-fixed tissues requires boiling tissue sections in 1mM EDTA buffer, pH 7.5-8.5, for 10-20 min followed by cooling at RT for 20 minutes). Researchers should empirically determine the suitability of the IGHD ighd for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "IgD (Immunoglobulin Delta Heavy Chain), Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
