Western blot (WB)
(Whole cell extracts (15ug protein) from untreated (-) or nocodazole-treated (+, 300ng/ml overnight) HeLa cells were resolved on a 4-12% Bis-Tris gel, transferred to a nitrocellulose membrane and probed with 1 ug/mL (1:500 dilution) of MBS6011004, overnight at 4°C. Proteins were visualized by chemiluminescence detection using IgG (HRP) goat anti-mouse antibody (1:3000). Direct-Blot™ anti-Beta-Actin was used as a loading control at a 1:25,000. Lane M: 'Molecular Weight Marker.)
Immunofluorescence (IF)
(HeLa cells were fixed with 4% paraformaldehyde (PFA) for 15 minutes, permeabilized with 0.5% Triton X-100 for 3 minutes, and blocked with 5% FBS for 60 minutes. Then the cells were intracellularly stained with MBS6011004 (1:2500) (0.2ug/ml) (lower) overnight at 4°C followed by Alexa Fluor® 594 (red) conjugated goat anti-rat IgG incubated for one hour at RT. Nuclei were counterstained with DAPI (blue). The image was captured with a 60X objective. The images displayed HeLa cells during cell cycle progression.)
Immunofluorescence (IF)
(Untreated HeLa (Panel A) and overnight nocodazoled-treated HeLa (Panel B) were stained with MBS6011004, followed by Alexa Fluor® 488 anti-alpha-tubulin, DyLight™ 594 goat anti-rat-lgG and DAPI.)

Variant histone H3 which replaces conventional H3 in a wide range of nucleosomes in active genes. Constitutes the predominant form of histone H3 in non-dividing cells and is incorporated into chromatin independently of DNA synthesis. Deposited at sites of nucleosomal displacement throughout transcribed genes, suggesting that it represents an epigenetic imprint of transcriptionally active chromatin. Nucleosomes wrap and compact DNA into chromatin, limiting DNA accessibility to the cellular machineries which require DNA as a template. Histones thereby play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. DNA accessibility is regulated via a complex set of post-translational modifications of histones, also called histone code, and nucleosome remodeling.
