Entrez Summary: Chromatin is a highly specialized structure composed of tightly compacted chromosomal DNA. Gene expression within the nucleus is controlled, in part, by a host of protein complexes which continuously pack and unpack the chromosomal DNA. One of the known mechanisms of this packing and unpacking process involves the acetylation and deacetylation of the histone proteins comprising the nucleosomal core. Acetylated histone proteins confer accessibility of the DNA template to the transcriptional machinery for expression. Histone deacetylases (HDACs) are chromatin remodeling factors that deacetylate histone proteins and thus, may act as transcriptional repressors. HDACs are classified by their sequence homology to the yeast HDACs and there are currently 2 classes. Class I proteins are related to Rpd3 and members of class II resemble Hda1p.HDAC4 is a class II histone deacetylase containing 1084 amino acid residues. HDAC4 has been shown to interact with NCoR. HDAC4 is a member of the class II mammalian histone deacetylases, which consists of 1084 amino acid residues. Its C terminal sequence is highly similar to the deacetylase domain of yeast HDA1. HDAC4, unlike other deacetylases, shuttles between the nucleus and cytoplasm in a process involving active nuclear export. Association of HDAC4 with 14-3-3 results in sequestration of HDAC4 protein in the cytoplasm. In the nucleus, HDAC4 associates with the myocyte enhancer factor MEF2A. Binding of HDAC4 to MEF2A results in the repression of MEF2A transcriptional activation. HDAC4 has also been shown to interact with other deacetylases such as HDAC3 as well as the corepressors NcoR and SMART.

NCBI Description

Histones play a critical role in transcriptional regulation, cell cycle progression, and developmental events. Histone acetylation/deacetylation alters chromosome structure and affects transcription factor access to DNA. The protein encoded by this gene belongs to class II of the histone deacetylase/acuc/apha family. It possesses histone deacetylase activity and represses transcription when tethered to a promoter. This protein does not bind DNA directly, but through transcription factors MEF2C and MEF2D. It seems to interact in a multiprotein complex with RbAp48 and HDAC3. [provided by RefSeq, Jul 2008]

Uniprot Description

HDAC4: a transcriptional regulator of the histone deacetylase family, subfamily 2. Deacetylates lysine residues on the N-terminal part of the core histones H2A, H2B, H3 AND H4. Plays an important role in transcriptional regulation, cell cycle progression and developmental events. Does not bind DNA directly, but through transcription factors MEF2C and MEF2D. It seems to interact in a multiprotein complex with RbAp48 and HDAC3.

Product Notes

The HDAC4 hdac4 (Catalog #AAA475081) is an Antibody produced from Mouse Source: Human and is intended for research purposes only. The product is available for immediate purchase. The Anti-HDAC4 (N-term) Mouse mAb reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's HDAC4 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunoprecipitation (IP). WB: 1:1000. Researchers should empirically determine the suitability of the HDAC4 hdac4 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "HDAC4, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
