• Hybridoma: Produced from a hybridoma resulting from the fusion of a mouse myeloma with B cells obtained from a rat immunized with purified, E. coli-derived recombinant human Hck (rhHck; aa 22-80; accession # P08631).
• Preparation and Storage: Lyophilized powder may be stored at 4 degree C for short-term only. Reconstitute to nominal volume by adding sterile PBS with 40-50% glycerol and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Western Blot (WB)

(Western Blot shows lysates of A20 mouse B cell lymphoma cell line, DA3 mouse myeloma cell line, Raji human Burkitt's lymphoma cell line, and Ramos human Burkitt's lymphoma cell line using MBS632623. PVDF membrane was probed with 1ug/ml of MBS632623 followed by HRP­ conjugated Anti­Rat IgG Secondary Antibody. A specific band was detected for Hck at ~59kD (as indicated). This experiment was conducted under reducing conditions.)

Hck expression is prominent in cells of myeloid lineage, particularly in granulocytes and monocytes, with protein levels increasing during terminal differentiation and macrophage activation. Hck contains a unique amino-terminal domain that is subject to lipid modifications, targeting this kinase to the plasma membrane.

Function: Non-receptor tyrosine-protein kinase found in hematopoietic cells that transmits signals from cell surface receptors and plays an important role in the regulation of innate immune responses, including neutrophil, monocyte, macrophage and mast cell functions, phagocytosis, cell survival and proliferation, cell adhesion and migration. Acts downstream of receptors that bind the Fc region of immunoglobulins, such as FCGR1A and FCGR2A, but also CSF3R, PLAUR, the receptors for IFNG, IL2, IL6 and IL8, and integrins, such as ITGB1 and ITGB2. During the phagocytic process, mediates mobilization of secretory lysosomes, degranulation, and activation of NADPH oxidase to bring about the respiratory burst. Plays a role in the release of inflammatory molecules. Promotes reorganization of the actin cytoskeleton and actin polymerization, formation of podosomes and cell protrusions. Inhibits TP73-mediated transcription activation and TP73-mediated apoptosis. Phosphorylates CBL in response to activation of immunoglobulin gamma Fc region receptors.
