Monoclonal antibody to detect Glutathione Reductase in human samples.

Background: Glutathione reductase (GR) is a member of pyridine nucleotide-disulfide oxidoreductases, which includes the closely related enzymes thioredoxin reductase, lipoamide dehydrogenase, trypanothione reductase and mercuric ion reductase. GR is a cytoplasmic flavoenzyme widely distributed in aerobic organisms. The dimeric protein is composed of two identical subunits, each containing 1 FAD and 1 redox-active disulfide/dithiol as components of the catalytic apparatus. It plays a role in maintaining glutathione (GSH) in its reduced form by catalyzing the reduction of glutathione disulfide (GSSG). In most eukaryotic cells, GR maintains the ratio of [GSH]/[GSSG] elevated, and participates in several vital functions such as the detoxification of reactive oxygen species as well as protein and DNA biosynthesis.

NCBI Description

This gene encodes a member of the class-I pyridine nucleotide-disulfide oxidoreductase family. This enzyme is a homodimeric flavoprotein. It is a central enzyme of cellular antioxidant defense, and reduces oxidized glutathione disulfide (GSSG) to the sulfhydryl form GSH, which is an important cellular antioxidant. Rare mutations in this gene result in hereditary glutathione reductase deficiency. Multiple alternatively spliced transcript variants encoding different isoforms have been found. Uniprot Description

GSR: a class-I pyridine nucleotide-disulfide oxidoreductase. Maintains high levels of reduced glutathione in the cytosol. Alternative initiation results in a mitochondrial and a cytoplasmic isoform.

Protein type: Oxidoreductase; EC 1.8.1.7; Other Amino Acids Metabolism - glutathione

Chromosomal Location of Human Ortholog: 8p21.1

Cellular Component: mitochondrial matrix; cytosol; external side of plasma membrane

Molecular Function: mercury (II) reductase activity; electron carrier activity; FAD binding; mercury ion binding; glutathione-disulfide reductase activity; NADP binding

Biological Process: response to reactive oxygen species; glutathione metabolic process; nucleobase, nucleoside and nucleotide interconversion; cell redox homeostasis; nucleobase, nucleoside and nucleotide metabolic process; detoxification of mercury ion

Product Notes

The GSR gsr (Catalog #AAA840126) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The Glutathione Reductase Antibody (Clone 2B3) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's Glutathione Reductase can be used in a range of immunoassay formats including, but not limited to, Immunoprecipitation (IP), ELISA (EIA), Western Blot (WB). IP: 1-2 ul, ELISA. Researchers should empirically determine the suitability of the GSR gsr for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Glutathione Reductase, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
