Application Notes
Immunoprecipitation: 5 ug immunoprecipitates rat FceRI alpha from 500 ug of biotinylated rat basophilic leukemia cell lysate. The immunoprecipitate was resolved by electrophoresis and transferred to nitrocellulose. Biotinylated protein bands wew visualized using NeutrAvidin labeled with HRP and and enhanced chemiluminesce detection system.

Optimal dilutions to be determined by the researcher.

Whole rat basophilic leukemia cells, positive for the rat high-affinity receptor recognizing immunoglobulin E (FceRI).
Preparation and Storage
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

The allergic reaction is mediated through the IgE high affinity receptor (FceRI). When a multivalent antigen is presented, a redistribution of the monomeric IgE-FceRI receptor complexes cause the degranulation of mast cells and basophils, resulting in the subsequent release of factors responsible for the allergic reaction. The FceRI receptor is a tetrameric complex, consisting of an a-chain, a b-chain and a dimeric g-chain. While the a-chain is primarily involved with the binding of IgE, the signal is transferred through the g subunit, which contains the immunoreceptor tyrosine activation motifs (ITAM) critical for initiating receptor mediated signal transduction through the Syk and Lyn tyrosine kinases. The FceRI receptor is able to engage and disengage the IgE, providing a versatile model for studying the function of kinase coupled receptors.

NCBI Description

The immunoglobulin epsilon receptor (IgE receptor) is the initiator of the allergic response. When two or more high-affinity IgE receptors are brought together by allergen-bound IgE molecules, mediators such as histamine that are responsible for allergy symptoms are released. This receptor is comprised of an alpha subunit, a beta subunit, and two gamma subunits. The protein encoded by this gene represents the alpha subunit. [provided by RefSeq, Aug 2011]

Function: Binds to the Fc region of immunoglobulins epsilon. High affinity receptor. Responsible for initiating the allergic response. Binding of allergen to receptor-bound IgE leads to cell activation and the release of mediators (such as histamine) responsible for the manifestations of allergy. The same receptor also induces the secretion of important lymphokines.

Subunit structure: Tetramer of an alpha chain, a beta chain, and two disulfide linked gamma chains.
