Subunit Structure
Part of a complex containing at least CDYL, REST, WIZ, SETB1, EHMT1 and EHMT2 (PubMed:19061646). During DNA replication, it is recruited by SETDB1 to form a S phase-specific complex that facilitates methylation of H3 Lys-9 during replication-coupled chromatin assembly and is at least composed of the CAF-1 subunit CHAF1A, MBD1 and SETDB1 (PubMed:15327775). Probably part of a corepressor complex containing ZNF304, TRIM28, SETDB1 and DNMT1 (PubMed:24623306). Interacts with TRIM28/TIF1B (PubMed:11959841). Interacts with ATF7IP and ATF7IP2; the interaction with ATF7IP is required to stimulate histone methyltransferase activity and facilitate the conversion of dimethylated to trimethylated H3 Lys-9 (PubMed:14536086, PubMed:15691849). Interacts with MBD1; interaction is abolished when MBD1 is sumoylated (PubMed:15327775, PubMed:17066076). Interacts with CBX1 and CBX5 (PubMed:15899859). Entrez Summary: This gene encodes a histone methyltransferase. The encoded enzyme catalyzes the reaction of S-adenosyl-L-methionine and histone L-lysine to produce S-adenosyl-L-homocysteine and histone N (6)-methyl-L-lysine. The encoded protein likely functions in transcriptional repression. Alternatively spliced transcript variants have been described.
UniPort Summary Function: Histone methyltransferase that specifically trimethylates Lys-9 of histone H3. H3 Lys-9 trimethylation represents a specific tag for epigenetic transcriptional repression by recruiting HP1 (CBX1, CBX3 and/or CBX5) proteins to methylated histones. Mainly functions in euchromatin regions, thereby playing a central role in the silencing of euchromatic genes. H3 Lys-9 trimethylation is coordinated with DNA methylation. Probably forms a complex with MBD1 and ATF7IP that represses transcription and couples DNA methylation and histone Lys-9 trimethylation. Its activity is dependent on MBD1 and is heritably maintained through DNA replication by being recruited by CAF-1. SETDB1 is targeted to histone H3 by TRIM28/TIF1B, a factor recruited by KRAB zinc-finger proteins. Probably forms a corepressor complex required for activated KRAS-mediated promoter hypermethylation and transcriptional silencing of tumor suppressor genes (TSGs) or other tumor-related genes in colorectal cancer (CRC) cells (PubMed:24623306). Also required to maintain a transcriptionally repressive state of genes in undifferentiated embryonic stem cells (ESCs) (PubMed:24623306). Associates at promoter regions of tumor suppressor genes (TSGs) leading to their gene silencing (PubMed:24623306). The SETDB1-TRIM28-ZNF274 complex may play a role in recruiting ATRX to the 3-exons of zinc-finger coding genes with atypical chromatin signatures to establish or maintain/protect H3K9me3 at these transcriptionally active regions (PubMed:27029610).

NCBI Description

This gene encodes a histone methyltransferase which regulates histone methylation, gene silencing, and transcriptional repression. This gene has been identified as a target for treatment in Huntington Disease, given that gene silencing and transcription dysfunction likely play a role in the disease pathogenesis. Alternatively spliced transcript variants of this gene have been described.[provided by RefSeq, Jun 2011]

Uniprot Description

SETDB1: a protein lysine methyltransferase that specifically trimethylates K9 of histone H3 (H3K9me3), a specific tag for epigenetic transcriptional repression by recruiting HP1 (CBX1, CBX3 and/or CBX5) proteins to methylated histones. Unlike SUV39H H3K9 methyltransferase, which functions mainly in heterochromatin regions such as pericentric heterochromatin, SETDB1 functions mainly in euchromatic regions, playing a central role in the silencing of euchromatic genes. H3K9me3 is coordinated with DNA methylation. Interacts with a variety of proteins, including transcription factors (ERG), histone deacetylases (HDAC1/2), DNA methyltransferases (DNMT3A/B) and transcriptional co-repressors (mSin3A/B, MBD1, KAP-1, the ATFa-associated modulator mAM). Its activity is dependent on MBD1 and is heritably maintained through DNA replication by being recruited by CAF-1. Contains Tudor and methyl-CpG-binding domains, which may coordinate binding to methylated histones and methylated DNA, respectively. Is targeted to histone H3 by TIF1B, a factor recruited by KRAB zinc-finger proteins. Recruited by DNMT3A to silenced promoters in cancer cells. May play a role in the pathogenesis of Huntington's disease, since levels of SETDB1 and H3K9me3 are both increased in diseased brains. Belongs to the histone-lysine methyltransferase family. Suvar3-9 subfamily. Three isoforms of the human protein are produced by alternative splicing.

Product Notes

The ESET setdb1 (Catalog #AAA476097) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The Anti-ESET Mouse mAb reacts with Human, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's ESET can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Researchers should empirically determine the suitability of the ESET setdb1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "ESET, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
