DNA adenine methylase (DAM) is an enzyme that methylates adenine residues in the 4-base pair DNA sequence, GATC. This methylation reaction has been shown to be crucial in a variety of cellular processes in bacteria. These include (i) the control of initiation of DNA replication (GATC occurs 11 times in the highly conserved bacterial origin of replication sequence); (ii) methyl-directed mismatch repair; (iii) control of transposition of certain transposable DNA elements; and (iv) control of gene expression. With regard to gene expression, the recent report by Heithoff et al. (Science; 284:967-970; 1999) showed that the deletion of the DAM gene rendered Salmonella typhimurium totally avirulent. Antibody can be used for Western blotting (1-5 µg/ml). Other applications not yet evaluated. Optimal concentration should be evaluated by serial dilutions. The antibody reacts with the recombinant protein as produced by E.coli. We have no data to support to its reactivity with wild type DAM protein.

NCBI Description

Dam is a base flipping enzyme. [More information is available at EcoGene: EG10204]. DNA adenine methyltransferase (Dam), which is responsible for methylation of nearly all GATC sequences in Escherichia coli , is encoded by the dam gene and occurs at about 130 molecules per rapidly growing cell . [More information is available at EcoCyc: EG10204].

Uniprot Description

Function: Methylates DNA within the sequence GATC and protects the DNA from cleavage by the restriction endonuclease MboI. Although it shares sequence specificity with a number of type II restriction endonucleases and methylases, it is thought to act in postreplication mismatch repair rather than as a part of a restriction modification system. May also play a role in DNA replication.

Catalytic activity: S-adenosyl-L-methionine + DNA adenine = S-adenosyl-L-homocysteine + DNA 6-methylaminopurine.

Sequence similarities: Belongs to the N(4)/N(6)-methyltransferase family.
