Specificity
This item specifically recognises a unique epitope shared by the intermediate filament keratins, cytokeratin 7 and cytokeratin 17, a 54kDa type II basic cytokeratin, and 46kDa type I acidic cytokeratin respectively. Cytokeratins are located in the intracytoplasmic cytoskeleton of epithelial tissues and their neoplasms, including the lungs, ovary and breast. Each keratin filament consists of at least one type I and one type II keratin chain, which then assemble to form a cytoskeletal network, extending from the surface of the nucleus to the cell membrane. The expression of a particular type of cytokeratin depends mainly on the type of epithelium and the development stage of the cell, including its corresponding malignant state. Immunohistochemical analysis of cytokeratin profiles is therefore a valuable tool for characterization, and this particular antibody could be useful for the evaluation of some stratified epithelia which express cytokeratin 17, but not cytokeratin 7. This item is reported as suitable for use in immunocytochemistry, see Bartek, J. et al. for details.

NCBI Description

This gene encodes the type I intermediate filament chain keratin 17, expressed in nail bed, hair follicle, sebaceous glands, and other epidermal appendages. Mutations in this gene lead to Jackson-Lawler type pachyonychia congenita and steatocystoma multiplex. [provided by RefSeq, Aug 2008]

Uniprot Description

K17: a type I cytoskeletal keratin. The keratins are intermediate filament proteins responsible for the structural integrity of epithelial cells and are subdivided into cytokeratins and hair keratins. There are two types of cytoskeletal and microfibrillar keratin: type I (acidic; 40-55 kDa) [K9 to K20] and type II (neutral to basic; 56-70 kDa) [K1 to K8]. Both a basic and an acidic keratin are required for filament assembly. Generally associates with K6.

Protein type: Cytoskeletal

Chromosomal Location of Human Ortholog: 17q21.2

Cellular Component: cytoplasm; intermediate filament

Molecular Function: MHC class II receptor activity; protein binding; structural constituent of cytoskeleton; MHC class II protein binding

Biological Process: positive regulation of hair follicle development; epidermis development; hair follicle morphogenesis; positive regulation of translation; keratinization; intermediate filament organization; signal transduction; positive regulation of cell growth

Product Notes

The KRT17 krt17 (Catalog #AAA214467) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The MOUSE ANTI HUMAN CYTOKERATIN 7/17 reacts with Bovine, Pig and may cross-react with other species as described in the data sheet. AAA Biotech's CYTOKERATIN 7/17 can be used in a range of immunoassay formats including, but not limited to, Immunoprecipitation (IP), Immunohistology Paraffin*, Western Blot (WB). Western Blot: This item detects bands of approximately 54kDa and 46kDa in HeLa cytoskeleton cell lysates under reducing conditions. Immunohistology - Paraffin: Minimum Dilution: 20; Maximum Dilution: 30ug/ml; Application Note: This product requires protein digestion pre-treatment of paraffin sections e.g. trypsin or pronase. Western Blotting: Minimum Dilution: 2; Maximum Dilution: 3ug/ml. Researchers should empirically determine the suitability of the KRT17 krt17 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CYTOKERATIN 7/17, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
