Application Notes
Suitable for Flow Cytometry.
Dilution: Flow Cytometry: 10ul labels 1x10e6. Validated using human peripheral blood monocytes.
Flow Cytometry Protocol: 1. Cells may be Fc-blocked with 1ug of human IgG/10e5 cells for 15 minutes at RT. Do not wash excess blocking IgG from this reaction.
2. Add10ul of C8299-01F to 1x10e6 cells and incubate for 30 minutes at RT.
3. Remove unbound antibody by washing the cells twice in Flow Cytometry Staining Buffer (F4568-10). Note that whole blood requires a RBC lysis step at this point using Flow Cytometry Human Lyse Buffer.
4. Resuspended cells in F4568-10 for flow cytometry analysis. As a control for this analysis, cells in a separate tube should be treated with PE-labeled mouse IgG1 antibody.

Recombinant corresponding to extracellular domain of human CX3CR1 expressed in E. coli.
Preparation and Storage
May be stored at 4 degree C before opening. DO NOT FREEZE! Stable at 4 degree C as an undiluted liquid. Stable for 12 months at 4 degree C. Freezing R-Phycoerythrin (PE) conjugates will result in a substantial loss of activity. PE conjugates are sensitive to light.

CX3CR1 is a 40kD seven transmembrane chemokine receptor that is expressed on several T cell subsets, monocytes, macrophages, microglia and epithelial cells. CX3CR1 binding to membrane bound or soluble CX3CR1/Fractalkine promotes inflammatory responses by inducing monocyte adhesion to endothelial cells and macrophage activation. CX3CR1 polymorphisms are associated with the development of chronic inflammatory disorders. Alternately spliced isoforms have extended N-terminal extracellular regions that increase the potency of CX3CR1 as a fusion coreceptor for HIV-1. Human CX3CR1 shares 82% amino acid sequence identity with mouse and rat CX3CR1.

NCBI Description

Fractalkine is a transmembrane protein and chemokine involved in the adhesion and migration of leukocytes. The protein encoded by this gene is a receptor for fractalkine. The encoded protein also is a coreceptor for HIV-1, and some variations in this gene lead to increased susceptibility to HIV-1 infection and rapid progression to AIDS. Four transcript variants encoding two different isoforms have been found for this gene. [provided by RefSeq]

CX3CR1: Receptor for the CX3C chemokine fractalkine and mediates both its adhesive and migratory functions. Acts as coreceptor with CD4 for HIV-1 virus envelope protein (in vitro). Isoform 2 and isoform 3 seem to be more potent HIV-1 coreceptors than isoform 1. Defects in CX3CR1 are a cause of susceptibility to age- related macular degeneration type 12 (ARMD12). ARMD12 is a form of age-related macular degeneration, a multifactorial eye disease and the most common cause of irreversible vision loss in the developed world. In most patients, the disease is manifest as ophthalmoscopically visible yellowish accumulations of protein and lipid that lie beneath the retinal pigment epithelium and within an elastin-containing structure known as Bruch membrane. Belongs to the G-protein coupled receptor 1 family. 3 isoforms of the human protein are produced by alternative splicing.
