Related Product Information for anti-CD44 antibody
anti-mouse CD44 monoclonal antibody reacts with all isoforms of CD44 (Pgp-1, Ly-24) a polymorphic glycoprotein which is broadly distributed on hematopoietic cells and a variety of nonhematopoietic cells 1,2. CD44 is a cell adhesion receptor and its primary ligand is hyaluronan 3. This antibody is suitable for use in flow cytometry. It has also been reported to have application for immunoprecipitation, detection of soluable CD44 with ELISA, IHC of frozen sections and paraffin sections, and complement depletion 1,2,3.

NCBI Description

The protein encoded by this gene is a cell-surface glycoprotein involved in cell-cell interactions, cell adhesion and migration. It is a receptor for hyaluronic acid (HA) and can also interact with other ligands, such as osteopontin, collagens, and matrix metalloproteinases (MMPs). This protein participates in a wide variety of cellular functions including lymphocyte activation, recirculation and homing, hematopoiesis, and tumor metastasis. Transcripts for this gene undergo complex alternative splicing that results in many functionally distinct isoforms, however, the full length nature of some of these variants has not been determined. Alternative splicing is the basis for the structural and functional diversity of this protein, and may be related to tumor metastasis. [provided by RefSeq, Jul 2008]

Uniprot Description

Function: Receptor for hyaluronic acid (HA). Mediates cell-cell and cell-matrix interactions through its affinity for HA, and possibly also through its affinity for other ligands such as osteopontin, collagens, and matrix metalloproteinases (MMPs). Adhesion with HA plays an important role in cell migration, tumor growth and progression. In cancer cells, may play an important role in invadopodia formation. Also involved in lymphocyte activation, recirculation and homing, and in hematopoiesis. Altered expression or dysfunction causes numerous pathogenic phenotypes. Great protein heterogeneity due to numerous alternative splicing and post-translational modification events. Ref.31

Subunit structure: Interacts with PKN2

By similarity. Interacts with HA, as well as other glycosaminoglycans, collagen, laminin, and fibronectin via its N-terminal segment. Interacts with ANK, the ERM proteins (VIL2, RDX and MSN), and NF2 via its C-terminal segment. Ref.41

Subcellular location: Membrane; Single-pass type I membrane protein. Note: Colocalizes with actin in membrane protrusions at wounding edges

By similarity.

Tissue specificity: Isoform 10 (epithelial isoform) is expressed by cells of epithelium and highly expressed by carcinomas. Expression is repressed in neuroblastoma cells.

Domain: The lectin-like LINK domain is responsible for hyaluronan binding

By similarity.

Post-translational modification: Proteolytically cleaved in the extracellular matrix by specific proteinases (possibly MMPs) in several cell lines and tumors. Ref.28N- and O-glycosylated. O-glycosylation contains more-or-less-sulfated chondroitin sulfate glycans, whose number may affect the accessibility of specific proteinases to their cleavage site(s). It is uncertain if O-glycosylation occurs on Thr-637 or Thr-638. Ref.28 Ref.39Phosphorylated; activation of PKC results in the dephosphorylation of Ser-706 (constitutive phosphorylation site), and the phosphorylation of Ser-672. Ref.26 Ref.27

Polymorphism: CD44 is responsible for the Indian blood group system. The molecular basis of the In(A)=In1/In(B)=In2 blood group antigens is a single variation in position 46; In(B), the most frequent allele, has Arg-46.

Sequence similarities: Contains 1 Link domain.

Product Notes

The CD44 cd44 (Catalog #AAA520463) is an Antibody produced from Rat and is intended for research purposes only. The product is available for immediate purchase. The Anti-Mouse CD44, FITC (Clone IM7.8.1) (rat IgG2b) reacts with Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's CD44 can be used in a range of immunoassay formats including, but not limited to, Immunohistochemistry (IHC). Researchers should empirically determine the suitability of the CD44 cd44 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CD44, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
