Application Notes
Flow Cytometry: This antibody can be used at 0.2 - 1 ug/10^6 cells.

Western blot: Use at a concentration range of 0.5-2 ug/mL, the detection limit for recombinant SCF is approximately 2-4 ng/lane under non-reducing conditions using compatible secondary reagents.

Dilution: A quick spin of the vial followed by reconstitution in H2O to a concentration of 1 mg/mL.
Host Note
This antibody was produced from a hybridoma resulting from the fusion of a mouse myeloma with B cells obtained from a mouse immunized with recombinant E. coli-derived Human SCF.
Preparation and Storage
The lyophilized antibody is stable for at least 1 year from date of receipt at -20°C.
Upon reconstitution, this antibody can be stored in working aliquots at 2°-8°C for one month, or at -20°C for six months without detectable loss of activity.

Avoid repeated freeze/thaw cycles.

The platelet glycoprotein IIb (αIIb; CD41) constitutes the alpha subunit of a highly expressed platelet surface integrin protein. CD41 is a protein composed of two subunits that interact with CD61 in the presence of calcium to form a functional adhesive protein receptor. Damage to blood vessels results in the release of a variety of intracellular mediators as well as initiation of the hemostatic cascade that in combination activate CD41/CD61 to bind to a variety of proteins including fibrinogen, fibronectin, von Willebrand factor and vitronectin.CD41/CD61 was once considered to be a platelet specific-receptor complex and has long been used as a phenotypic marker for the megakaryocyte- platelet lineage.

NCBI Description

ITGA2B encodes integrin alpha chain 2b. Integrins are heterodimeric integral membrane proteins composed of an alpha chain and a beta chain. Alpha chain 2b undergoes post-translational cleavage to yield disulfide-linked light and heavy chains that join with beta 3 to form a fibronectin receptor expressed in platelets that plays a crucial role in coagulation. Mutations that interfere with this role result in thrombasthenia. In addition to adhesion, integrins are known to participate in cell-surface mediated signalling. [provided by RefSeq, Jul 2008]

ITGA2B: Integrin alpha-IIb/beta-3 is a receptor for fibronectin, fibrinogen, plasminogen, prothrombin, thrombospondin and vitronectin. It recognizes the sequence R-G-D in a wide array of ligands. It recognizes the sequence H-H-L-G-G-G-A-K-Q-A-G-D-V in fibrinogen gamma chain. Following activation integrin alpha- IIb/beta-3 brings about platelet/platelet interaction through binding of soluble fibrinogen. This step leads to rapid platelet aggregation which physically plugs ruptured endothelial cell surface. Belongs to the integrin alpha chain family. Heterodimer of an alpha and a beta subunit. The alpha subunit is composed of an heavy and a light chain linked by a disulfide bond. Alpha-IIb associates with beta-3. Directly interacts with RNF181. 3 isoforms of the human protein are produced by alternative splicing.
