CD4/L3T4 is a type I transmembrane glycoprotein expressed on most thymocytes and the helper/inducer subpopulation of mature T lymphocytes of all mouse strains tested.^1,2 CD4 has also been detected at low density on pluripotent hematopoietic stem cells, bone marrow myeloid precursors, and intrathymic lymphoid precursors. ^3-6 The CD4 antigen functions as a co-receptor, interacting with class II major histocompatability complex (MHC) molecules in the recognition of foreign antigens by T cells. ^1,2 In vivo and in vitro treatment with the GK1.5 mAb effectively depletes CD4^+ cells.^7,8 The mAb also blocks helper T cell responses to MHC class II antigens, including cytolysis, proliferation, allogeneic B cell help, and release of lymphokines.^1,8-13

Characterization
To ensure lot-to-lot consistency, each batch of monoclonal antibody is tested by flow cytometry to conform to characteristics of a standard reference reagent. Representative data are included in this package insert.
Preparation and Storage
The purified (UNLB) antibody is supplied as 0.5 mg of purified immunoglobulin in 1.0 mL of 100 mM borate buffered saline, pH 8.2. No preservatives or amine-containing buffer salts added. Store at 2-8 degree C.
The fluorescein (FITC) conjugate is supplied as 0.5 mg or 0.1 mg in 1.0 mL of PBS/NaN3. Store at 2-8 degree C.
The biotin (BIOT) conjugate is supplied as 0.5 mg in 1.0 mL of PBS/NaN3. Store at 2-8 degree C.
The R-phycoerythrin (R-PE) conjugate is supplied as 0.1 mg in 1.0 mL or 0.2 mg in 2.0 mL of PBS/NaN3 and a stabilizing agent. Store at 2-8 degree C. Do not freeze!
The allophycocyanin (APC) conjugate is supplied as as 0.1 mg in 1.0 mL of PBS/NaN3 and a stabilizing agent. Store at 2-8 degree C. Do not freeze!
The Spectral Red (SPRD), R-phycoerythrin -Texas Red (R-PE-TXRD), R-phycoerythrin-Cyanine 7 (RPE- CY 7), R-phycoerythrin-Cyanine 5.5 (R-PE-CY 5.5 Allophycocyanin-Cyanine 5.5 (APC-CY 5.5), and Allophycocyanin- Cyanine 7 (APC-CY 7) conjugates are supplied as 0.1 mg in 1.0 mL of PBS/NaN3 and a stabilizing agent. Store at 2-8 degree C. Do not freeze!
The low endotoxin, azide-free (LE/AF) antibody is supplied as 0.5 mg purified immunoglobulin in 1.0 mL of PBS. Aliquot and store at or below -20 degree C.
The Cyanine 5 (CY 5) conjugate is supplied as 0.1 mg in 1.0 mL of PBS/NaN3. Store at 2-8 degree C.
Protect conjugated forms from light. Aliquot and freeze the low endotoxin, azide-free product at -20 degree C immediately upon receipt. Each reagent is stable for the period shown on the bottle label if stored as directed.
