Host
Mouse
Reactivity
Human
Clonality
Monoclonal
Isotype
Mouse IgG1 kappa
Clone Number
SN6
Specificity
Antibody SN6 recognizes an extracellular epitope of the human CD105 homodimer with a molecular weight of about 160 kd. It has been reported to cross react with non human Primate as well as Equine and Porcine (11) CD105.
Form/Format
50mM Sodium Phosphate pH 7.5, 500mM Potassium Chloride, 150mM NaCl, 15% Glycerol, 0.2%BSA, 0.04% NaN3 (as a preservative).
Concentration
Volume in Vial: 0.2 ml (varies by lot)
Applicable Applications for anti-CD105 antibody
Flow Cytometry (FC/FACS) (12), Immunohistochemistry (IHC)
Application Notes
Working Dilution: 1:50 (or use 1.6ul of concentrated stock per 5 x 105-cell test)
Immunogen
Membrane preparation of human B-lineage leukemia cells
Conjugation
Allophycocyanin (APC)
Production
Protein A purified antibody from tissue culture supernatant was conjugated to cross-linked Allophycocyanin through a sulfo-ester linkage. Unconjugated antibody was removed using size exclusion chromatography. Antibody conjugate is at 0.5mg/ml with an A650/A280 ratioof 1.96.
Performance
Five x 105 cultured human Nalm-6cells per tube were washed and incubated 45 minutes on ice with 80ul of anti-CD105/APC at a dilution factor of 1:50 (10ug/ml). Cells were washed three times, fixed and analyzed byFACS. Cells stained positive with a mean shift of 1.55 log10 fluorescent units when compared to a Mouse IgG1/APC negative control (#MBS666633). Binding was blocked when cells were preincubated 10 minutes with 20ul of 0.5mg/ml anti-CD105 antibody (#MBS666489).
Preparation and Storage
Store at 2-5 degree C. Do not freeze! Protect from light.
Product should retain activity for at least 12 months after shipping date when stored as recommended.

Related Product Information for anti-CD105 antibody
Human CD105 (endoglin) is a homodimeric membrane glycoprotein (1) virtually restricted to endothelial cells, and present on some leukemia cells and minor subsets of bone marrow cells. CD105 binds to TGF-b1 and b3 with high affinity (4). Antibody SN6 will augment binding of TGF-b1 to CD105 expressing leukemia cells. Antibody SN6 has been used to identify mesenchymal progenitor cells populations (8,9,10)

Product Notes

The CD105 (Catalog #AAA666951) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The Monoclonal anti-human CD105 (Endoglin)/APC reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's CD105 can be used in a range of immunoassay formats including, but not limited to, Flow Cytometry (FC/FACS) (12), Immunohistochemistry (IHC). Working Dilution: 1:50 (or use 1.6ul of concentrated stock per 5 x 105-cell test). Researchers should empirically determine the suitability of the CD105 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CD105, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
