Application Notes
Suitable for use in ELISA and Western Blot.
Dilution: Western Blot: 1-2ug/ml with the appropriate secondary reagents to detect mouse CDO. Using a colorimetric detection system, the detection limit for rmCDO is approximately 5 ng/lane under non-reducing and reducing conditions. Chemiluminescent detection will increase sensitivity by 5 to 50 fold.
ELISA (direct): 0.5-1.0ug/ml with the appropriate secondary reagents to detect mCDO. The detection limit for rmCDO is approximately 1ng/well.

NS0-derived rmCDO extracellular domain.
Preparation and Storage
Lyophilized powder may be stored at 4 degree C for short-term only. Reconstitute to nominal volume by adding sterile 40-50% glycerol, PBS and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Mouse CDO is a 190kD member of the Ig/FN-type III repeat subfamily of transmembrane proteins. The extracellular region contains five Ig-like domains and three FN-III modules. The molecule forms cis-complexes with itself and the related molecule termed BOC. Mouse CDO extracellular domain shares 95% and 85% aa identity with the corresponding regions of rat and human CDO, respectively.

Uniprot Description

CDON: Component of a cell-surface receptor complex that mediates cell-cell interactions between muscle precursor cells. Promotes differentiation of myogenic cells. Defects in CDON are the cause of holoprosencephaly type 11 (HPE11). HPE11 is a structural anomaly of the brain, in which the developing forebrain fails to correctly separate into right and left hemispheres. Holoprosencephaly is genetically heterogeneous and associated with several distinct facies and phenotypic variability. 2 isoforms of the human protein are produced by alternative splicing.
