This antibody specifically recognizes an epitope within Abetax-42. NOTE: When administered to young Tg2576 mice with minimal Abeta deposition, this antibody reduced Abeta accumulation in the brain.

Application Notes
mmunoblotting, Immunohistochemistry: Im m unofluorescence, Immunoprecip i tation Test at 1-10ug/ml. Sandwich ELISA (as capture antibody with #MBS190935 as HRP - c onjugated detection antibody). These are recommended concentrations; enduser should determine optimal concentrations for their applications. Sandwich ELISA protoco l on next page. Sandwich ELISA Protocol:
96-well ELISA plates are coated with capture MAbat 2.5-5ug/well and incubated at 4 degree C overnight. The next day 300ul of blocking buffer is added, and plates are again incubated at 4 degree C overnight. The next day plates are washed in PBS and serial dilution of A beta samples are added; plates are incubated overnight at 4 degree C. The next day plates are washed in PBS, and HRP-conjugated detection Abeta MAb is added; plates are incubated for 4hrs at room temperature. Plates are washed in PBS-Tween and developed with TMB substrate for 5 mins

Dilution Instructions
Dilute in PBS or medium which is identical to that used in the assay system.
Preparation and Storage
This antibody is stable for at least one (1) year at -20 degree C.

Accumulation and aggregation of amyloid beta (Abeta) in the brain is indicated as the trigger of a pathological cascade that causes Alzheimer disease (AD). The highly amyloidogenic 42-amino acid form of Abeta (Abeta42) and aminoterminally truncated forms of Abeta (Abetax-42) are the predominant species of Abeta typically found in diffuse and senile plaques within the AD brain. The relative levels of Abeta42 appear to be the key regulators of Abeta aggregation into amyloid; thus Abeta42 has been implicated as the initiating molecule in the pathogenesis of AD.

Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Amyloid Beta 1-42, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
