Western Blot (WB)
(Western Blot Positive WB detected in: L02 whole cell lysate, K562 whole cell lysate, Hela whole cell lysate, Raji whole cell lysate, A549 whole cell lysate All lanes: ACLY antibody at 1:1500 Secondary Goat polyclonal to rabbit IgG at 1/50000 dilution Predicted band size: 121, 120, 92 kDa Observed band size: 120 kDa)
Immunofluorescence (IF)
(Immunofluorescence staining of HepG2 Cells at 1?50, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeated by 0.2% TritonX-100, and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4 degree C. Nuclear DNA was labeled in blue with DAPI. The secondary antibody was FITC-conjugated AffiniPure Goat Anti-Rabbit IgG ?H+L?.)
Flow Cytometry (FC/FACS)
(Overlay histogram showing Hela cells stained with (red line) at 1?50. The cells were fixed with 70% Ethylalcohol (18h) and then incubated in 10% normal goat serum to block non-specific protein-protein interactions followedby the antibody (1ug/1*106cells) for 1 h at 4?.The secondary antibody used was FITC-conjugated goat anti-rabbit IgG (H+L) at 1/200 dilution for 30min at 4?. Control antibody (green line) was Rabbit IgG (1ug/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.)
NCBI and Uniprot Product Information
NCBI Description
ATP citrate lyase is the primary enzyme responsible for the synthesis of cytosolic acetyl-CoA in many tissues. The enzyme is a tetramer (relative molecular weight approximately 440,000) of apparently identical subunits. It catalyzes the formation of acetyl-CoA and oxaloacetate from citrate and CoA with a concomitant hydrolysis of ATP to ADP and phosphate. The product, acetyl-CoA, serves several important biosynthetic pathways, including lipogenesis and cholesterogenesis. In nervous tissue, ATP citrate-lyase may be involved in the biosynthesis of acetylcholine. Two transcript variants encoding distinct isoforms have been identified for this gene. Uniprot Description
ACLY: ATP citrate-lyase is the primary enzyme responsible for the synthesis of cytosolic acetyl-CoA in many tissues. Has a central role in de novo lipid synthesis. In nervous tissue it may be involved in the biosynthesis of acetylcholine. Homotetramer. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes
The ACLY acly (Catalog #AAA7136725) is a Recombinant Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The ACLY Antibody reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's ACLY can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA), Western Blot (WB), Immunofluorescence (IF), Flow Cytometry (FC/FACS). WB: 1:500-1:5000 IF: 1:20-1:200 FC/FACS: 1:20-1:200. Researchers should empirically determine the suitability of the ACLY acly for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "ACLY, Monoclonal Recombinant Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
