Immunohistochemistry (IHC) (Figure 2. IHC analysis of ACACA using anti-ACACA antibody (M01802).ACACA was detected in paraffin-embedded section. Heat mediated antigen retrieval was performed in citrate buffer (pH6, epitope retrieval solution) for 20 mins. The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1ug/ml rabbit anti-ACACA Antibody (M01802) overnight at 4° C. Biotinylated goat anti Rabbit IgG IgG antibody was used as secondary antibody and incubated for 30 minutes at 37° C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC) with DAB as the chromogen. Figure 1. Western blot analysis of ACACA using anti-ACACA antibody (M01802).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel)/90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-ACACA antigen affinity purified polyclonal antibody at 0.5 ug/mL overnight at 4° C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-Rabbit IgG IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for ACACA. Previous Next )

Immunogen
A synthesized peptide derived from human Acetyl-CoA Carboxylase ACC1 a subunit of acetyl-CoA carboxylase (ACC), a multifunctional enzyme system. Catalyzes the carboxylation of acetyl-CoA to malonyl-CoA, the rate-limiting step in fatty acid synthesis. Acetyl-CoA carboxylase (ACC) catalyzes the pivotal step of the fatty acid synthesis pathway. The 265 kDa ACC Alpha (ACC1) is the predominant isoform found in liver, adipocytes, and mammary gland, while the 280 kDa ACC Beta (ACC2) is the major isoform in skeletal muscle and heart.
Preparation and Storage
Store at 4 degree C short term. Store at -20 degree C long term
Avoid freeze/thaw cycles.

NCBI Description

Acetyl-CoA carboxylase (ACC) is a complex multifunctional enzyme system. ACC is a biotin-containing enzyme which catalyzes the carboxylation of acetyl-CoA to malonyl-CoA, the rate-limiting step in fatty acid synthesis. There are two ACC forms, alpha and beta, encoded by two different genes. ACC-alpha is highly enriched in lipogenic tissues. The enzyme is under long term control at the transcriptional and translational levels and under short term regulation by the phosphorylation/dephosphorylation of targeted serine residues and by allosteric transformation by citrate or palmitoyl-CoA. Multiple alternatively spliced transcript variants divergent in the 5' sequence and encoding distinct isoforms have been found for this gene. [provided by RefSeq, Jul 2008]

Uniprot Description

Catalyzes the rate-limiting reaction in the biogenesis of long-chain fatty acids. Carries out three functions: biotin carboxyl carrier protein, biotin carboxylase and carboxyltransferase.

Product Notes

The ACACA acaca (Catalog #AAA1752138) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-Acetyl-CoA Carboxylase Monoclonal Antibody reacts with Human, Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's Acetyl-CoA Carboxylase can be used in a range of immunoassay formats including, but not limited to, Immunohistochemistry (IHC), Western Blot (WB). WB: 1:1000-1:2000 IHC: 1:50-1:200. Researchers should empirically determine the suitability of the ACACA acaca for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Acetyl-CoA Carboxylase, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
