Preparation and Storage
This product is shipped at ambient temperature. It is recommended to aliquot and store at -20 degree C on receipt. When thawed, aliquot the sample as needed. Keep aliquots at 2-8 degree C for short term use (up to 4 weeks) and store the remaining aliquots at -20 degree C.

Avoid repeated freezing and thawing as this may denature the antibody. Storage in frost-free freezers is not recommended.

Mouse anti Human A2M antibody, clone M5, recognizes alpha-2-macroglobulin (A2M), also known as CPAMD5. A2M is a major proteinase inhibitor, with the ability to bind most proteinases and many growth factors, hormones and cytokines in human blood and tissue (Kurz et al. 2017). A2M is largely produced by the liver under conditions of stress, and is then secreted into the blood and extracellular spaces. It is also produced locally by macrophages, fibroblasts, and epithelial cells (Kim et al. 2017).

A2M levels dramatically decrease as humans age, a process which has been hypothesized to facilitate tumor development. In naked mole rats, a species with pronounced resistance to cancer, A2M has been shown to inhibit tumor growth regardless of tumor origin (Kurz et al. 2017).

The biotinylated Mouse anti Human A2M antibody, clone M5 can be used as a detection antibody in a sandwich ELISA with the purified Mouse anti Human A2M antibody, clone M17 as the capture antibody.

NCBI Description

Alpha-2-macroglobulin is a protease inhibitor and cytokine transporter. It inhibits many proteases, including trypsin, thrombin and collagenase. A2M is implicated in Alzheimer disease (AD) due to its ability to mediate the clearance and degradation of A-beta, the major component of beta-amyloid deposits. [provided by RefSeq, Jul 2008]

Uniprot Description

A2M: Is able to inhibit all four classes of proteinases by a unique 'trapping' mechanism. This protein has a peptide stretch, called the 'bait region' which contains specific cleavage sites for different proteinases. When a proteinase cleaves the bait region, a conformational change is induced in the protein which traps the proteinase. The entrapped enzyme remains active against low molecular weight substrates (activity against high molecular weight substrates is greatly reduced). Following cleavage in the bait region a thioester bond is hydrolyzed and mediates the covalent binding of the protein to the proteinase. Belongs to the protease inhibitor I39 (alpha-2- macroglobulin) family.
