Specificity
Recognizes human alpha-skeletal and alpha-cardiac (sarcomeric) actins as well as neoplasms derived from normal tissues (i.e. rhabdomyosarcomas). Shows no crossreaction with smooth muscle actin. Species Crossreactivity: bovine, rabbit, sheep, rat, guinea pig and frog.

Application Notes
Suitable for use in Immunofluorescence and Immunohistochemistry.
Dilution: Immunohistochemistry (formalin/paraffin sections): 1:200 for 20 min at RT. No special pretreatment is required for the immunohistochemical staining of formalin/paraffin tissues.
Immunogen
N-terminal decapeptide of alpha skeletal muscle isoform of actin; acetylated at the N-terminus. Cellular Localization: Cytoplasmic. Epitope: N-terminal decapeptide.

Skeletal or Cardiac muscle, or Rhabdomyosarcoma
Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile 40-50% glycerol, aliquot and store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Uniprot Description

Function: Actins are highly conserved proteins that are involved in various types of cell motility and are ubiquitously expressed in all eukaryotic cells.

Subunit structure: Polymerization of globular actin (G-actin) leads to a structural filament (F-actin) in the form of a two-stranded helix. Each actin can bind to 4 others.

Subcellular location: Cytoplasm › cytoskeleton.

Post-translational modification: Oxidation of Met-46 and Met-49 by MICALs (MICAL1, MICAL2 or MICAL3) to form methionine sulfoxide promotes actin filament depolymerization. MICAL1 and MICAL2 produce the (R)-S-oxide form. The (R)-S-oxide form is reverted by MSRB1 and MSRB2, which promote actin repolymerization

Miscellaneous: In vertebrates 3 main groups of actin isoforms, alpha, beta and gamma have been identified. The alpha actins are found in muscle tissues and are a major constituent of the contractile apparatus. The beta and gamma actins coexist in most cell types as components of the cytoskeleton and as mediators of internal cell motility.
