Monkey CD27 antigen (CD27) ELISA Kit
Principle of the Assay: CD27 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CD27 antibody and an CD27-HRP conjugate. The assay sample and buffer are incubated together with CD27-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CD27 concentration since CD27 from samples and CD27-HRP conjugate compete for the anti-CD27 antibody binding site. Since the number of sites is limited, as more sites are occupied by CD27 from the sample, fewer sites are left to bind CD27-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CD27 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a member of the TNF-receptor superfamily. This receptor is required for generation and long-term maintenance of T cell immunity. It binds to ligand CD70, and plays a key role in regulating B-cell activation and immunoglobulin synthesis. This receptor transduces signals that lead to the activation of NF-kappaB and MAPK8/JNK. Adaptor proteins TRAF2 and TRAF5 have been shown to mediate the signaling process of this receptor. CD27-binding protein (SIVA), a proapoptotic protein, can bind to this receptor and is thought to play an important role in the apoptosis induced by this receptor. [provided by RefSeq, Jul 2008]
