Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for MMP-10 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MMP-10 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MMP-10 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MMP-10 bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

Proteins of the matrix metalloproteinase (MMP) family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. Most MMP's are secreted as inactive proproteins which are activated when cleaved by extracellular proteinases. The enzyme encoded by this gene degrades proteoglycans and fibronectin. The gene is part of a cluster of MMP genes which localize to chromosome 11q22.3. [provided by RefSeq, Jul 2008]

Uniprot Description

Function: Can degrade fibronectin, gelatins of type I, III, IV, and V; weakly collagens III, IV, and V. Activates procollagenase.

Catalytic activity: Similar to stromelysin 1, but action on collagen types III, IV and V is weak.1 PublicationManual assertion based on experiment in:Ref.8

Cofactor: Binds 2 zinc ions per subunit.1 PublicationManual assertion based on experiment in:Ref.8

Product Notes

The Human MMP10 mmp10 (Catalog #AAA703295) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA703295 ELISA Kit recognizes Human MMP10. It is sometimes possible for the material contained within the vial of "matrix metallopeptidase 10 (stromelysin 2), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
