Principle of the Assay: MIP-3beta ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-MIP-3beta antibody and an MIP-3beta-HRP conjugate. The assay sample and buffer are incubated together with MIP-3beta-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MIP-3beta concentration since MIP-3beta from samples and MIP-3beta-HRP conjugate compete for the anti-MIP-3beta antibody binding site. Since the number of sites is limited, as more sites are occupied by MIP-3beta from the sample, fewer sites are left to bind MIP-3beta-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MIP-3beta concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene is one of several CC cytokine genes clustered on the p-arm of chromosome 9. Cytokines are a family of secreted proteins involved in immunoregulatory and inflammatory processes. The CC cytokines are proteins characterized by two adjacent cysteines. The cytokine encoded by this gene may play a role in normal lymphocyte recirculation and homing. It also plays an important role in trafficking of T cells in thymus, and in T cell and B cell migration to secondary lymphoid organs. It specifically binds to chemokine receptor CCR7.
