Principle of the assay: MAPK3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-MAPK3 antibody and an MAPK3-HRP conjugate. The assay sample and buffer are incubated together with MAPK3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MAPK3 concentration since MAPK3 from samples and MAPK3-HRP conjugate compete for the anti-MAPK3 antibody binding site. Since the number of sites is limited, as more sites are occupied by MAPK3 from the sample, fewer sites are left to bind MAPK3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MAPK3 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the Ser/Thr protein kinase family. This kinase functions as a mitogen-activated protein kinase (MAP kinase)- activated protein kinase. MAP kinases are also known as extracellular signal-regulated kinases (ERKs), act as an integration point for multiple biochemical signals. This kinase was shown to be activated by growth inducers and stress stimulation of cells. In vitro studies demonstrated that ERK, p38 MAP kinase and Jun N-terminal kinase were all able to phosphorylate and activate this kinase, which suggested the role of this kinase as an integrative element of signaling in both mitogen and stress responses. This kinase was reported to interact with, phosphorylate and repress the activity of E47, which is a basic helix-loop-helix transcription factor known to be involved in the regulation of tissue-specific gene expression and cell differentiation.
