Principle of the Assay||This kit was based on sandwich enzyme-linked immune-sorbent assay technology. anti-Human LTbeta antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-Human LTbeta antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Human LTbeta amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of Human LTbeta can be calculated.

NCBI Description

Lymphotoxin beta is a type II membrane protein of the TNF family. It anchors lymphotoxin-alpha to the cell surface through heterotrimer formation. The predominant form on the lymphocyte surface is the lymphotoxin-alpha 1/beta 2 complex (e.g. 1 molecule alpha/2 molecules beta) and this complex is the primary ligand for the lymphotoxin-beta receptor. The minor complex is lymphotoxin-alpha 2/beta 1. LTB is an inducer of the inflammatory response system and involved in normal development of lymphoid tissue. Lymphotoxin-beta isoform b is unable to complex with lymphotoxin-alpha suggesting a function for lymphotoxin-beta which is independent of lympyhotoxin-alpha. Alternative splicing results in multiple transcript variants encoding different isoforms. [provided by RefSeq, Jul 2008]

Isoform 2 is probably non-functional. Belongs to the tumor necrosis factor family. 2 isoforms of the human protein are produced by alternative splicing.
