Intended Uses: This KLKB1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rat KLKB1. This ELISA kit for research use only!

Principle of the Assay: KLKB1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-KLKB1 antibody and an KLKB1-HRP conjugate. The assay sample and buffer are incubated together with KLKB1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the KLKB1 concentration since KLKB1 from samples and KLKB1-HRP conjugate compete for the anti-KLKB1 antibody binding site. Since the number of sites is limited, as more sites are occupied by KLKB1 from the sample, fewer sites are left to bind KLKB1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The KLKB1 concentration in each sample is interpolated from this standard curve.

NCBI Description

The import of proteins into the nucleus is a process that involves at least 2 steps. The first is an energy-independent docking of the protein to the nuclear envelope and the second is an energy-dependent translocation through the nuclear pore complex. Imported proteins require a nuclear localization sequence (NLS) which generally consists of a short region of basic amino acids or 2 such regions spaced about 10 amino acids apart. Proteins involved in the first step of nuclear import have been identified in different systems. These include the Xenopus protein importin and its yeast homolog, SRP1 (a suppressor of certain temperature-sensitive mutations of RNA polymerase I in Saccharomyces cerevisiae), which bind to the NLS. KPNA2 protein interacts with the NLSs of DNA helicase Q1 and SV40 T antigen and may be involved in the nuclear transport of proteins. KPNA2 also may play a role in V(D)J recombination [provided by RefSeq, Jul 2008]

Uniprot Description

KPNA2: Functions in nuclear protein import as an adapter protein for nuclear receptor KPNB1. Binds specifically and directly to substrates containing either a simple or bipartite NLS motif. Docking of the importin/substrate complex to the nuclear pore complex (NPC) is mediated by KPNB1 through binding to nucleoporin FxFG repeats and the complex is subsequently translocated through the pore by an energy requiring, Ran- dependent mechanism. At the nucleoplasmic side of the NPC, Ran binds to importin-beta and the three components separate and importin-alpha and -beta are re-exported from the nucleus to the cytoplasm where GTP hydrolysis releases Ran from importin. The directionality of nuclear import is thought to be conferred by an asymmetric distribution of the GTP- and GDP-bound forms of Ran between the cytoplasm and nucleus. Belongs to the importin alpha family.

Product Notes

The Canine KPNa2 kpna2 (Catalog #AAA7231777) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7231777 ELISA Kit recognizes Canine KPNa2. It is sometimes possible for the material contained within the vial of "Karyopherin Alpha 2, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
