INTENDED USE This KLK8 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Chicken KLK8. This ELISA kit for research use only!

PRINCIPLE OF THE ASSAY KLK8 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-KLK8 antibody and an KLK8-HRP conjugate. The assay sample and buffer are incubated together with KLK8-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the KLK8 concentration since KLK8 from samples and KLK8-HRP conjugate compete for the anti-KLK8 antibody binding site. Since the number of sites is limited, as more sites are occupied by KLK8 from the sample, fewer sites are left to bind KLK8-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The KLK8 concentration in each sample is interpolated from this standard curve.

NCBI Description

Kallikreins are a subgroup of serine proteases having diverse physiological functions. Growing evidence suggests that many kallikreins are implicated in carcinogenesis and some have potential as novel cancer and other disease biomarkers. This gene is one of the fifteen kallikrein subfamily members located in a cluster on chromosome 19. Alternate splicing of this gene results in four transcript variants encoding four different isoforms. The isoforms exhibit distinct patterns of expression that suggest roles in brain plasticity and ovarian cancer. [provided by RefSeq, Jul 2008]

KLK8: Serine protease which is capable of degrading a number of proteins such as casein, fibrinogen, kininogen, fibronectin and collagen type IV. Also cleaves L1CAM in response to increased neural activity. Induces neurite outgrowth and fasciculation of cultured hippocampal neurons. Plays a role in the formation and maturation of orphan and small synaptic boutons in the Schaffer- collateral pathway, regulates Schaffer-collateral long-term potentiation in the hippocampus and is required for memory acquisition and synaptic plasticity. Involved in skin desquamation and keratinocyte proliferation. Plays a role in the secondary phase of pathogenesis following spinal cord injury. Belongs to the peptidase S1 family. Kallikrein subfamily. 4 isoforms of the human protein are produced by alternative splicing.
