Western Blot (WB)

(Recombinant KAT7 protein activity assay 0.5 ug Histone H4 was incubated with 0 ug (-), 0.4 ug (+) KAT7 in 20 ul reaction system for 2 hours at room temperature. Half of each reaction was run on a 13% SDS-PAGE gel, and products were detected by Western blot. H4ac (pan-acetyl) antibody was used to recognize acetylated histone H4.)

Background: KAT7 (K(Lysine) Acetyltransferase 7) is a MYST-family histone acetyltransferase (HAT) that forms a complex with ING4/5, JADE1/2/3, and Eaf6, which is found enriched on coding regions of genes, supporting a role in transcriptional elongation. In addition, KAT7 is thought to participate in events controlling the initiation of DNA replication.

Uniprot Description

MYST2: a histone acetyltransferase that participates in DNA replication and the loading of the minichromosome maintenance (Mcm) complex onto chromatin. Is required for origin recognition complex (Orc) formation and replication licensing, and is responsible for much of the acetylation of histone H4. Interacts with MCM2 and ORC1L. Phosphorylated by Cdc2 during mitosis, creating a site that recruits Plk1, which in turn phosphorylates MYST2. Its phosphorylation by Plk1 during mitosis may be required for the formation of the prereplicative complex. Interacts with the androgen receptor (AR) in the presence of dihydrotestosterone. Specifically represses AR-mediated transcription.
