NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between sIL-2alphaR and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This sIL-2alphaR ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Monkey sIL-2alphaR. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: sIL-2alphaR ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-sIL-2alphaR antibody and an sIL-2alphaR-HRP conjugate. The assay sample and buffer are incubated together with sIL-2alphaR-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the sIL-2alphaR concentration since sIL-2alphaR from samples and sIL-2alphaR-HRP conjugate compete for the anti-sIL-2alphaR antibody binding site. Since the number of sites is limited, as more sites are occupied by sIL-2alphaR from the sample, fewer sites are left to bind sIL-2alphaR-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The sIL-2alphaR concentration in each sample is interpolated from this standard curve.

NCBI Description

The interleukin 2 (IL2) receptor alpha (IL2RA) and beta (IL2RB) chains, together with the common gamma chain (IL2RG), constitute the high-affinity IL2 receptor. Homodimeric alpha chains (IL2RA) result in low-affinity receptor, while homodimeric beta (IL2RB) chains produce a medium-affinity receptor. Normally an integral-membrane protein, soluble IL2RA has been isolated and determined to result from extracellular proteolyisis. Alternately-spliced IL2RA mRNAs have been isolated, but the significance of each is presently unknown. Mutations in this gene are associated with interleukin 2 receptor alpha deficiency.[provided by RefSeq, Nov 2009]
