Principle of the Assay: IL-29 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-IL-29 antibody and an IL-29-HRP conjugate. The assay sample and buffer are incubated together with IL-29-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the IL-29 concentration since IL-29 from samples and IL-29-HRP conjugate compete for the anti-IL-29 antibody binding site. Since the number of sites is limited, as more sites are occupied by IL-29 from the sample, fewer sites are left to bind IL-29-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The IL-29 concentration in each sample is interpolated from this standard curve.

This gene encodes a cytokine distantly related to type I interferons and the IL-10 family. This gene, interleukin 28A (IL28A), and interleukin 28B (IL28B) are three closely related cytokine genes that form a cytokine gene cluster on a chromosomal region mapped to 19q13. Expression of the cytokines encoded by the three genes can be induced by viral infection. All three cytokines have been shown to interact with a heterodimeric class II cytokine receptor that consists of interleukin 10 receptor, beta (IL10RB) and interleukin 28 receptor, alpha (IL28RA).
