Intended Uses: This IL-1beta ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Porcine IL-1beta. This ELISA kit for research use only!

Principle of the Assay: IL-1beta ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-IL-1beta antibody and an IL-1beta-HRP conjugate. The assay sample and buffer are incubated together with IL-1beta-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the IL-1beta concentration since IL-1beta from samples and IL-1beta-HRP conjugate compete for the anti-IL-1beta antibody binding site. Since the number of sites is limited, as more sites are occupied by IL-1beta from the sample, fewer sites are left to bind IL-1beta-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The IL-1beta concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a member of the interleukin 1 cytokine family. This cytokine is produced by activated macrophages as a proprotein, which is proteolytically processed to its active form by caspase 1 (CASP1/ICE). This cytokine is an important mediator of the inflammatory response, and is involved in a variety of cellular activities, including cell proliferation, differentiation, and apoptosis. The induction of cyclooxygenase-2 (PTGS2/COX2) by this cytokine in the central nervous system (CNS) is found to contribute to inflammatory pain hypersensitivity. This gene and eight other interleukin 1 family genes form a cytokine gene cluster on chromosome 2. [provided by RefSeq, Jul 2008]

Uniprot Description

IL1B: Produced by activated macrophages, IL-1 stimulates thymocyte proliferation by inducing IL-2 release, B-cell maturation and proliferation, and fibroblast growth factor activity. IL-1 proteins are involved in the inflammatory response, being identified as endogenous pyrogens, and are reported to stimulate the release of prostaglandin and collagenase from synovial cells. Monomer. Belongs to the IL-1 family.

Product Notes

The Rat IL-1beta il1b (Catalog #AAA732184) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA732184 ELISA Kit recognizes Rat IL-1beta. It is sometimes possible for the material contained within the vial of "Interleukin 1beta, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
