Principle of the Assay: IL-13 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-IL-13 antibody and an IL-13-HRP conjugate. The assay sample and buffer are incubated together with IL-13-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the IL-13 concentration since IL-13 from samples and IL-13-HRP conjugate compete for the anti-IL-13 antibody binding site. Since the number of sites is limited, as more sites are occupied by IL-13 from the sample, fewer sites are left to bind IL-13-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The IL-13 concentration in each sample is interpolated from this standard curve.

This gene encodes an immunoregulatory cytokine produced primarily by activated Th2 cells. This cytokine is involved in several stages of B-cell maturation and differentiation. It up-regulates CD23 and MHC class II expression, and promotes IgE isotype switching of B cells. This cytokine down-regulates macrophage activity, thereby inhibits the production of pro-inflammatory cytokines and chemokines. This cytokine is found to be critical to the pathogenesis of allergen-induced asthma but operates through mechanisms independent of IgE and eosinophils.
