Preparation and Storage
Lyophilized and reconstituted products are stable for 6 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Furher dilutions can be made in assay buffer.

DDC (EC 1.1.128) belongs to the family group II Decarboxylase (DDC, GAD, HDC and TYRD). DDC catalyzes the formation of tryptamine from L-tyrosine. It also acts on 5-hydroxy-LO-tryptophan and dihydroxy-L-Phenylalanine (DOPA). Human DDC (480aa, chromosome 7p11) is a homodimer in the native state. it requires pyridoxal phosphate as a cofactor.

NCBI Description

The encoded protein catalyzes the decarboxylation of L-3,4-dihydroxyphenylalanine (DOPA) to dopamine, L-5-hydroxytryptophan to serotonin and L-tryptophan to tryptamine. Defects in this gene are the cause of aromatic L-amino-acid decarboxylase deficiency (AADCD). AADCD deficiency is an inborn error in neurotransmitter metabolism that leads to combined serotonin and catecholamine deficiency. Multiple alternatively spliced transcript variants encoding different isoforms have been identified for this gene. [provided by RefSeq, Jun 2011]

Function: Catalyzes the decarboxylation of L-3,4-dihydroxyphenylalanine (DOPA) to dopamine, L-5-hydroxytryptophan to serotonin and L-tryptophan to tryptamine.

Catalytic activity: L-dopa = dopamine + CO2.5-hydroxy-L-tryptophan = 5-hydroxytryptamine + CO2.

Cofactor: Pyridoxal phosphate. Ref.10

Pathway: Catecholamine biosynthesis; dopamine biosynthesis; dopamine from L-tyrosine: step 2/2.

Involvement in disease: Aromatic L-amino-acid decarboxylase deficiency (AADCD) [MIM:608643]: An inborn error in neurotransmitter metabolism that leads to combined serotonin and catecholamine deficiency. It causes developmental and psychomotor delay, poor feeding, lethargy, ptosis, intermittent hypothermia, gastrointestinal disturbances. The onset is early in infancy and inheritance is autosomal recessive.Note: The disease is caused by mutations affecting the gene represented in this entry. Ref.3 Ref.11 Ref.14 Ref.15
