iNOS Phospho-Regulation
Background: Nitric oxide (NO) has a broad range of biological activities and is implicated in many cell signaling pathways. Nitric oxidesynthases (NOS), the enzymes responsible for synthesis of NO, are fused enzymes that include a cytochrome reductaseand a cytochrome that requires three cosubstrates (L-arginine, NADPH, and oxygen) and five cofactors (FAD, FMN, calmodulin, tetrahydrobiopterin, and heme). The NOS family includes two constitutive Ca2+/CaM-dependent forms ofNOS: nNOS (also designated bNOS, NOS-I), whose activity was first identified in neurons and eNOS (also designatedecNOS, NOS-III) first identified in endothelial cells. The inducible form of NOS, iNOS (also designated NOS-II), is Ca2+independent and is induced by microbial products in various cell types. Phosphorylation of NOS enzymes may regualtetheir activity. For example, Src kinase-induced phosphorylation of Tyr-1055 in iNOS reduces proteasomal degradation ofiNOS, while shear stress induced phosphorylation of Tyr-657 in eNOS attenuates enzyme activity. Both of these tyrosinesites are conserved in nNOS, and may have important roles for regulate neuronal NOS activity.

Product Notes
The Human, Rat, Mouse iNOS (Catalog #AAA474464) is an Antibody Sampler Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA474464 Antibody Sampler Kit recognizes Human, Rat, Mouse iNOS. AAA Biotech's iNOS Phospho-Regulation can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), ELISA (EIA), Immunocytochemistry (ICC). Researchers should empirically determine the suitability of the iNOS for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "iNOS Phospho-Regulation, Antibody Sampler Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
